谷红注射液抗脑缺血性再灌注损伤的作用及其机制 被引量:19

摘要 为探讨谷红注射液抗脑缺血再灌注损伤的作用及机制,本实验将SD大鼠随机分为假手术组、模型组、谷红注射液低、中、高剂量组、尼莫地平注射液组,每组10只。在建立大鼠大脑中动脉局灶性栓塞(MCAO)模型的基础上,观察大鼠神经功能缺损症状,脑梗死体积比,各组大鼠脑组织中超氧化物歧化酶(SOD)活力,丙二醛(MDA)含量,谷胱甘肽过氧化物酶(GSH-Px)和过氧化氢酶(CAT)的活性;以及血清组织型纤溶酶原激活物(t-PA)、纤溶酶原激活物抑制因子(PAI)含量、血栓素B2(TXB2)和6-酮前列腺素F1α(6-keto-PGF1α)的含量。结果表明谷红注射液能显著促进神经功能缺损症状的恢复,缩小大鼠手术后的脑梗死体积,显著提高脑组织中SOD,GSH-Px和CAT活性,降低MDA含量,同时还可显著提高血清中t-PA的含量,降低PAI含量,降低TXB2的含量,提高6-keto-PGF1α含量。说明谷红注射液具有较好的抗氧化和抗血栓的作用,对大鼠脑缺血再灌注损伤有明显的保护作用。 To explore the effect and mechanism of Guhong injection against cerebral ischemia reperfusion injury, SD rats were randomly divided into the sham-operated group, the model group, the nimodipine group, and high, medium and low-dose Guhong injection groups, with 10 rats in each group. The middle cerebral artery embolization (MCAO) model was established to observe neuro- logical deficit symptoms, infarct volume, SOD activity, MDA content, GSH-Px and CAT activity in rats, as well as the contents of t-PA, PAI, TXB2 and 6-keto-PGF1αin serum. The results showed that Guhong injection could obviously promote the recovery of neurological deficit symptoms, narrow the brain infarct volume in rats after surgery, significantly increase the activities of SOD, GSH-Px and CAT and decrease the content of MDA. Meanwhile, it also could obviously increase the contents of t-PA and 6-keto-PGFl, and decrease the contents of PAI and TXB2 in serum, indicating that Guhong injection have better anfioxidant and anfithrombus effects, as well as a significant protective effect against cerebral ischemia reperfusion injury in rats.
