摘要
目的:研究喉癌组织中脆性组氨酸三联体基因(fragile histidine traid,FHIT)和基质金属蛋白酶-2(matrix metalloproteinase-2,MMP-2)的表达以及与临床病理特征的关系。方法:采用免疫组化链酶卵白素-生物素复合体(strep avidin-biotin complex,SABC)法检测FHIT、MMP-2蛋白在92例喉鳞状细胞癌及相距10 mm癌旁组织和25例正常喉腔黏膜中的表达。结果:FHIT蛋白在喉癌组织中的阳性率(43.48%)明显低于癌旁组织(86.96%)及喉正常黏膜组织(92.00%),组间比较,差异均有统计学意义(P<0.05),FHIT蛋白的表达与临床分期和淋巴结转移有关,而与年龄、分化程度无关;MMP-2在喉癌组织中的阳性率(79.35%)明显高于癌旁组织(18.48%)及喉正常黏膜组织(12.00%),经比较,差异均有统计学意义(P<0.05),MMP-2的表达与分化程度和临床分期有关,而与年龄、淋巴结转移无关;此外,FHIT与MMP-2表达呈负相关。结论:FHIT、MMP-2的异常表达与喉癌的发生、发展及预后有关,二者在喉癌的发生、发展中起协同作用。
Objective :To explore and analyze the expression of fragile histidine traid (FHIT )and matrix metalloprotei_nase 2(MMP -2) protein in laryngocarcinoma and their relationship with clinicopathological .Methods :Expression of FHIT and MMP-2 protein was measured with immunohistochemistry method in 92 cases of laryngeal squamous cell car_cinoma(LSCC) tissue ,adjacent mucosa tissue which were 1 .0 cm away from carcinoma and 25 cases of normal laryngeal mucosa .Results :The expression of FHIT positive in laryngocarcinoma was 43 .48% significantly less than that adjacent to carcinoma(86 .96% ) and normal laryngeal mucosa (92% ) .There was a significant difference ( P〈0 .05) .FHIT expres_sion level was correlated with clinical stages and lymphnode metastasis ,and has no correlation with age and differentiation level .The expression of MMP-2 positive in laryngocarcinoma was 79 .35% significantly higher than that adjacent to car_cinoma (18 .48% ) and normal laryngeal mucosa (12% ) .There was a significant difference ( P 〈0 .05) .MMP -2 ex_pression level was correlated with clinical stages and differentiation level ,has no correlation with age and lymphnode me_tastasis ;FHIT was negatively correlated with MMP -2 .Conclusion:Experimental results show that the abnormal ex_pression of FHIT and MMP-2 may play an important role in the occurrence ,development and prognosis of laryngocarci_noma .They may play a synergistic role in laryngeal cancer occurrence and development .
