摘要
为建立准确、灵敏的UPLC-MS/MS方法直接测定止咳平喘类中药制剂及保健食品中非法添加的茶碱的含量,采用Agilent ZORBAX-SB-C18柱为分析柱,以甲醇-0.1%甲酸铵(30:70)为流动相,检测波长270nm,扫描方式采用全扫描一级质谱、全扫描二级质谱以及MRM模式定量,质量数范围100~300。结果显示UPLC-MS/MS方法测定茶碱的检出限为scan模式下50ng/ml、MRM模式下0.05ng/ml;茶碱的定量限为MRM模式下0.1ng/ml,在0.1ng/ml^2.0ng/ml范围内浓度与峰面积呈良好的线性关系(r=0.9975)。该方法建立的系统能够灵敏、准确地测定止咳平喘类中药制剂及保健食品中茶碱含量。
To establish a rapid, accurate UPLC -MS/MS method for the determination of theophylline, which were illegally added into traditional Chinese medicines and healthcare products. Method: The analytes were separated on a Agilent ZORBAX-SB-C18 column with methanol -0.1% ammonium formate (30:70) as mobile phase, detecting wavelength at 270nm, in three scan modes including full scan MS, full scan MS2, and MRM model. Results: A good resolution was obtained for theophylline under the UPLC and MS conditions. The determination limits were in the range of 0. lng/ml - 2.0ng/ml. Conclusion: The method is simple, rapid, accurate and highly sensitive, which can be used for the determination of traditional Chinese medicine and healthcare products mixed with theophylline.
