摘要
采用响应面法对热浸法提取流苏石斛多糖提取工艺进行优化,并以清除1,1-二苯基-2-三硝基苯肼(DPPH)自由基能力为评价指标,对不同产地流苏石斛多糖抗氧化活性进行比较研究。结果表明:流苏石斛多糖提取最佳工艺参数为提取温度92℃、料液比(V/W)26∶1、提取时间2.5 h,理论提取率为9.31%,实际提取率为9.18%,相对误差为1.40%;兴义、罗甸、云南、广西4个产地的流苏石斛多糖含量分别为10.34%、8.36%、3.35%、6.66%;以IC50作为判定抗氧化活性强弱的指标,分析表明,4个不同产地中以云南产流苏石斛多糖抗氧化能力相对较强。
The extraction technology of polysaccharides from Dendrobiumfimbriatum by reflux method was optimized by using the response surface method. The antioxidant activities of D. fimbriatum polysaccharides collected from different regions in China were investigated using in vitro antioxidant assays: 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities. The results showed that the optimal extraction conditions were as follows: extraction temperature of 92%, ratio of material to solvent of 1:26 and extraction time of 2.5 h. Under these conditions, the yield of D. fimbriatum polysaccharides was up to 9.18%, the best predicative extraction rate of the polysaccharides was 9.31%, and the relative error was 1.40%. The contents of polysaccharides in D. fimbriatum from Xingyi Guizhou, Luodian Guizhou, Yunnan, Guangxi were 10.34%, 8.36%, 3.35% and 6.66%, respectively. Among these 4 samples, the sample collected from Yunnan had the relatively higher antioxidant activity by using ICs0 value as an index.
