摘要
文章利用紫外可见光谱和荧光光谱研究了生理条件下呋喃唑酮与牛血红蛋白(BHb)的相互作用.实验结果表明:呋喃唑酮分子与牛血红蛋白发生反应生成基态复合物,导致BHb内源荧光猝灭,该猝灭属于静态猝灭.测定了279.15 K,292.15 K温度下该反应的结合位点数,结合常数(K)和结合热力学参数.K值分别为1.884×105L/mol-1和1.714×105L/mol-1,标准焓变和熵变分别为-8.25 Kjmol-1和-0.83 jmol-1K-1.热力学参数的变化表明呋喃唑酮与BHb之间以氢键或范德华作用力为主.
The interaction between furazolidone and Bovine Hemoglobin(BHb) was studied using UV/Vis and fluorescence spectroscopy.The experimental results showed that the quenching of BHb fluorescence by furazolidone was a static one and was attributed to the formation of furazolidone-BHb complex.The binging constants,K,at 279.15 K and 292.15 K,were obtained,with the values as 1.884* 10^5Lmol^-1,and 1.714* 10^5Lmol^-1,respectively.The standard enthalpy chang(ΔH^θ) and the standard entropy(ΔS^θ) were-8.25 Kjmol^-1and-0.83 Jmol^-1K^-1respectively.The hydrogen bonding and vander waals force played a major role in stabilizing the complex.The effect of furazolidone on the conformation of BHb was analyzed with synchronous fluorescence spectroscopy.
