抗原修复时间对陈旧性乳腺癌核抗原表达的影响

Impact of Antigen Retrieval Time on the Expression of Nuclear Antigen in Old Breast Cancer Paraffin

目的探索使陈旧性乳腺癌石蜡标本ER、PR、Ki-67在免疫组化检测中获得高表达的抗原修复时间。方法采用Envision二步法,将120例存放>2年的乳腺癌石蜡组织切片,分别在EDTA(p H9.0)修复液中高压修复5、10、15、20min和水浴修复30min,比较抗原表达差异。结果 ER、PR在修复时间>15min时阳性率明显提高,Ki-67在高压修复15、20min组阳性率较水浴30min、高压修复5、10min组明显提高。ER、PR、Ki-67在高压修复20min时阳性信号强,阳性率最高,与修复5、10min组差异有显著性(P<0.05)。结论高压修复20min可显著提高陈旧性乳腺癌石蜡组织ER、PR、Ki-67抗原的检出率。

Objective To explore the time of antigen retrieval with high expression for immunohistochemical staining of ER、PR、Ki-67 in old breast cancer paraffin. Methods Immunohistochemical staining of 120 old breast cancer paraffin cases stored for over 2 years was processed by Envision method and each tissue was retrieved in EDTA（pH9.0）antigen retrieval buffer under high pressure for 5, 10, 15 and 20 minutes respectively and water-bathed for 30 minutes;the antigen expressions were compared. Results The positive rates of ER and PR were obviously increased when the retrieval time was over 15 minutes;Ki-67 was of a higher positive rate when re-trieved under high-pressure for 15 and 20 minutes than when water-bathed for 30minutes and when retrieved under high pressure for 5 and 10 minutes;ER, Pr and Ki-67 showed strong positive signals and the highest positive rates when retrieved under high pressure for 20 minutes, significantly different from the positive rates when retrieved for 5 and 10 minutes（P〈0.05）. Conclusions Retrieval under high pressure for 20 minutes can obviously raise the detection rate of ER, PR and Ki-67 in old breast cancer paraffin.
