摘要
目的:研究运动对肥胖大鼠脂肪细胞线粒体含量、活性及有氧代谢相关基因的影响。方法:采用普利莱法测定线粒体含量、活性,采用分光光度法测定细胞色素氧化酶(COX)的活性,采用RT-PCR测定有氧代谢相关基因细胞色素C(Cytc)、肉碱棕榈酰转移酶1(CPT1)、COX和苹果酸脱氢酶MDH mRNA的表达水平。结果:与对照组大鼠相比,肥胖组大鼠脂肪细胞线粒体的含量、活性显著降低,线粒体呼吸链上的限速酶COX活性以及有氧代谢相关基因Cytc、CPT1、COX和MDH mRNA表达显著降低;与肥胖组大鼠相比,运动组大鼠脂肪细胞线粒体的含量、活性显著升高;线粒体呼吸链上的限速酶COX活性以及有氧代谢相关基因Cytc、CPT1、COX和MDH mRNA表达显著升高。结论:有氧运动可能通过提高脂肪细胞线粒体含量、活性和改善有氧氧化呼吸功能而发挥减重作用。
Objective: This paper aims to study the effect of exercise on adipocytes mitochondrial content, activity and aerobic metabolism related genes of obese rats. Methods: pulilai method is adopted to test mitochondrial content, activity; spectrophotometry is adopted to test the COX activity; RT-PCR is adopted to test the expression levels of aerobic metabolism related genes Cytc, carnitine palmitoyl transferaseI, COX, MDH mRNA. Results: Compared with rats in the control group, the obese group has a significant decrease in the mitochondrial fatty cells as well as expression levels of aerobic metabolism related genes Cytc, carnitine palmitoyl transferaseI, COX, MDH mRNA. While compared with obese rates, the exercise intervention group has a significant increase in the mitochondrial fatty cells as well as expression levels of aerobic metabolism related genes Cytc, carnitine palmitoyl transferaseI, COX, MDH mRNA. Conclusion: the aerobic exercise may increase adipocytes mitochondrial content, activity, and improve the aerobic oxidation of respiratory function for losing weight.
