补肾、健脾、活血法对骨质疏松症小鼠骨及骨骼肌中Na^+-K^+-ATP酶含量的影响 被引量:5

Effects of tonifying kidney, invigorating the spleen and promoting blood circulation on the content of Na^+-K^+-ATP enzyme in bone and skeletal muscle of mice with osteoporosis

摘要 目的:观察补肾、健脾、活血法对骨质疏松症(OP)小鼠骨骼及骨骼肌Na+-K+-ATP酶含量的影响,探讨中医防治OP的作用机制。方法:小鼠分为正常组、模型组、补肾组、健脾组、活血组和福善美组。灌胃8周,取股骨及骨骼肌,采用ELISA方法测定Na+-K+-ATP酶的含量。结果:1与正常组比较,其他各组小鼠骨骼肌中Na+-K+-ATP酶降低(P<0.01);与模型组比较,补肾组、福善美组小鼠骨骼肌Na+-K+-ATP酶升高(P<0.01);健脾组和活血组与模型组比较无统计学意义。2与正常组比较,其他各组小鼠骨Na+-K+-ATP酶降低(P<0.01);与模型组比较,补肾组、福善美组小鼠骨Na+-K+-ATP酶升高(P<0.01);健脾组和活血组与模型组比较无统计学意义。结论:OP的形成可能与骨及骨骼肌中Na+-K+-ATP酶含量异常有关;补肾法通过调控OP小鼠的骨及骨骼肌中Na+-K+-ATP酶含量,对OP具有一定的防治作用。 Objective: Observation of the effects of tonifying kidney, invigorating the spleen and promoting blood circulation on the content of Na^+-K^+-ATP enzyme in bone and skeletal muscle of mice with osteoporosis, to explore its mechanism. Methods: Mice were randomly divided into normal group, model group, tonifying kidney group, invigorating spleen group, promoting blood circulation group and Fosamax group. Expect for the normal group and model group, all groups received corresponding medicine by gavage for 8 weeks. Then, the content of Na^+-K^+-ATP enzyme in bone and skeletal muscle was determined with ELISA method. Results: (1)Compared with the normal group, the content of Na^+-K^+-ATP enzyme in skeletal muscle in the other groups significantly decreased (P〈0.01). Compared with the model group, the content of Na^+-K^+-ATP enzyme in skeletal muscle in the tonifying kidney group and Fosamax group significantly increased (P〈0.01). Although the content of Na^+-K^+-ATP enzyme in skeletal muscle in invigorating spleen group and promoting blood circulation group increased as well, there were no difference in both groups above compared with the model group. (2)Compared with the normal group, the content of Na^+-K^+-ATP enzyme in bone in the other groups significantly decreased (P〈0.01). Compared with the model group, the content of Na^+-K^+-ATP enzyme in the tonifying kidney group and Fosamax group significantly increased (P〈0.01). Although the content of Na^+-K^+-ATP enzyme in bone invigorating spleen group and the promoting blood circulation group increased as well, there was no difference in 3 groups above compared with the model group. Conclusion: Osteoporosis may be related to the abnormality of content of Na^+-K^+-ATP enzyme in bone and skeletal muscle. The methods of tonifying have certain preventive effect on osteoporosis, the mechanism of which may be that tonifying kidney and invigorating spleen could regulate the content of Na^+-K^+-ATP enzyme in bone and skeletal muscle.
