摘要
目的:探讨中药清毒栓治疗宫颈高危型人乳头瘤病毒(HPV)感染的分子机制。方法:以HPV18阳性的宫颈癌细胞系He La为细胞模型,清毒栓水提醇沉液、β-榄香烯、干扰素α-2b进行干预;分别采用Western blot及Real-time PCR检测主要组织相容性复合物-Ⅰ(MHC-Ⅰ)抗原呈递通路相关分子TAP1、TAP2、LMP2、LMP7的蛋白及基因表达差异。结果:与对照组相比,清毒栓及β-榄香烯均可显著上调He La细胞内抗原呈递通路相关分子TAP1、TAP2、LMP2、LMP7的蛋白表达(P<0.05),清毒栓可上调4种分子的基因表达(P<0.05),β-榄香烯仅能可上调TAP2的基因表达(P<0.05),干扰素α-2b对该4种分子的基因表达无影响。结论:中药清毒栓可通过调节抗原呈递通路相关分子的蛋白及基因表达,起到促进抗原呈递的作用,这可能是该方疗效的作用机制之一。
Objective: To explore the molecular mechanism of Qingdu suppository (QDS) in the treatment of high risk HPV infection. Methods: Human cervical cancer cell line HeLa as the model (HPV 18 positive) was cultured in vitro and treated by the extraction of QDS, β-elemene and IFN-α-2b. The protein and gene expression of MHC- I antigen presentation pathway associated molecules including TAP1, TAP2, LMP2 and LMP7 were detected by Western blot and Real-time PCR. Results: Compared with the control group, the treatment drugs could increase the protein expression of antigen presentation pathway associated molecules TAP1, TAP2, LMP2 and LMP7 in cell line HeLa (P〈0.05). QDS could up-regulate the mRNA expression of four molecules (P〈0.05), β -elemene could only up-regulate the gene expression of TAP2 (P〈0.05), while there was no significant difference after treatment with IFN-α -2b. Conclusion: QDS could promote antigen presentation through up-regulating the gene and protein expression of antigen presentation associated molecules. This might be one of the mechanisms of QDS effects. β -elemene and IFN α -2b should affect post transcriptional events to promote protein expression of antigen presentation associated molecules, ultimately promoted antigen presentation of infected cells.
