秦艽抗肝损伤活性部位筛选 被引量:11

Screening of active part of Gentiana macrophylla Pall. in the liver protection

摘要 目的:筛选秦艽发挥保肝作用的有效部位。方法:用系统溶剂法对秦艽95%乙醇提取物进行萃取分离,分成4个极性部位,用所得不同极性部位的药液灌胃大鼠7d后,皮下注射CCl4花生油溶液,致急性肝损伤大鼠模型,测定血清学指标(ALT,AST)并制作病理学切片,观察肝组织病理改变。结果:与模型组比较,秦艽提取物水溶性部位的高、低剂量组均可明显抑制大鼠血清中ALT、AST的升高(P<0.01);病理学切片显示,秦艽提取物水部位能明显减轻模型动物肝脏病变程度。结论:水溶性部位为秦艽保肝护肝的活性部位。 Objective: To screen the effective fraction with liver protection activity from Gentiana macrophylla Pail. Methods: Fractions of Gentiana macrophylla Pail. with different polarities were obtained from a serious of extraction solutions. The activities of AST and ALT were measured in liver injury mouse model induced by carbon tetrachloride (CCl4). Results: With the system by method of solvent extraction separation of ethanol extract of Gentiana 95%, divided into 4 polar fractions, with the different polar parts of liquid gastric perfusion rat 7 days later, subcutaneous injection of CCl4 peanut oil solution, induced acute liver injury in a rat model, determination of serological indexes (ALT, AST) and the production of pathological sections, observation of pathological changes of liver tissue. Conclusion: Compared with the model group, Gentiana macrophylla extract water at high, low dose group can significantly inhibit ALT in rat serum, the increase of AST (P〈0.01); pathological biopsy showed, Gentiana macrophylla extract water position can significantly reduce the severity of the liver lesions in animal model.
