摘要
探索纤维素酶法提取制备复方半边莲注射液的最佳工艺条件,采用单因素试验和正交试验设计,运用纤维素酶法提取制备复方半边莲注射液,并以HPLC法检测指标成分野黄芩苷含量为指标。结果表明,各因素的影响次序为酶解温度、p H、酶用量、酶解时间,最佳工艺条件为酶解温度45℃,酶用量为7.2 mg/g,最适p H为5.0,酶解时间为60 min。在此条件下,复方半边莲注射液中野黄芩苷含量为2.42mg/m L,明显大于注册标准中煎煮法野黄芩苷含量1.34 mg/m L。相对原注册标准煎煮法,纤维素酶法提取制备复方半边莲注射液工艺简单可行,具有提取效率高、生产周期短等优点。
The conditions of extracting compound lobelia injection with cellulase were optimized.The scutellarin of compound lobelia injection was extracted by cellulase and determined by HPLC in orthogonal experiment.The optimal conditions were the enzymolysis temperature of 45 ℃,the pH of 5.0,the enzymolysis time of 60 minutes.Under these conditions,the content of scutellarin in compound lobelia injection was 2.42 mg/mL,significantly more than 1.34 mg/mL through decocting technology contained in registration standards.Cellulase extraction of compound lobelia injection is simple and feasible,with advantages of high efficiency and short production duration.
