摘要
以三色堇(Viola tricolor L.)叶片为材料,采用SDS法、CTAB法及改良CTAB法分别对三色堇叶片DNA进行了提取,并用琼脂糖凝胶电泳和紫外光谱对DNA提取质量和浓度进行检测。结果表明,改良CTAB法提取的DNA纯度最高,提取效率较高;SDS法提取的DNA含有较多的蛋白质或糖类杂质;CTAB法虽然能够除去蛋白质等杂质,但是DNA浓度低于改良CTAB法。
High-quality DNA sample is important for plant molecular study.DNA was extracted from Viola tricolor L.leaves with SDS,CTAB and modified CTAB.The quality and concentration of extracted DNA was detected by agarose gel electrophoresis and ultraviolet spectrum.The results showed that the purity of DNA extracted with modified CTAB method was the best.DNA extracted with SDS method contained more impurities including protein or carbohydrate.Although CTAB method removed proteins,DNA concentration was lower than that with the modified CTAB method.The modified CTAB method was an appropriate,rapid and practical way for extracting DNA from Viola tricolor L.leaves.
