Effects of the lipid nanoparticles on the general toxicity and endocrine system in mice

摘要 目的研究脂质纳米粒(LNs)对小鼠的全身毒性及内分泌系统的影响。方法小鼠尾静脉注射LNs,连续7 d,继续培养23 d后,于眼眶取血,分离血清,用免疫试剂盒法检测血清中皮质醇(COR)、生长激素(GH)、睾酮(T)和甲状腺素(T4)的含量;并剖离小鼠组织(心、肝、脾、肺、肾和脑)行病理检查。结果同正常组小鼠相比,给予LNs后小鼠的各组织未见病理学变化,其内分泌系统也无太大影响(P>0.05)。结论 LNs对小鼠全身无明显毒性且不影响内分泌系统,可应用于静脉注射。 OBJECTIVE To observe the effects of the lipid nanoparticles on the general toxicity and the endocrine system in mice.METHODS Mice were subjected to iv injection of LNs for 7 days. And after 23 days housing,mice were sacrificed and the serum was collected. And then the cortisol( COR),growth hormone( GH),testosterone( T) and thyroxine( T4) were measured. The tissues( Heart,Liver,Spleen,Lung,Kidney and Brain) were collected and fixed by formalin. And then the pathology of tissues were assessed after HE staining. RESULTS Compared with the normal mice,the LNs mice had no changes in the pathology of tissues and LNs showed a little influence on the parameters of the endocrine system( P 〉 0. 05). CONCLUSION LNs show a consderable margin of safety in vivo and no influence on the endocrine system of mice,suggesting that LNs can be used as an injectable carrier in vivo.
