Determination of patchoulic alcohol and pogostone in the rat plasma by GC-MS

摘要 目的采用GC-MS法同时测定大鼠血浆中广藿香醇和广藿香酮的含量。方法采用Agilent HP-5ms色谱柱(30 m×0.25 mm,0.25μm),电子碰撞离子源(EI),电子倍增电压(EMV)模式,检测离子分别为:广藿香醇m/z 222.2、广藿香酮m/z 168.0。结果广藿香醇5.040-2016 ng·m L^-1(r=0.9979)、广藿香酮1.020-406.0 ng·m L^-1(r=0.9958)与峰面积比值的线性关系良好;批内、批间的方法回收率均〈±15%,批内、批间精密度的RSD均〈8%;平均萃取回收率分别为76.4%、80.6%。结论所用方法简单、灵敏、特异性高、定量准确,可为大鼠血浆中广藿香醇和广藿香酮的药动学研究提供参考。 OBJECTIVE To establish a method for the determination of patchoulic alcohol and pogostone in the rat plasma by GC-MS. METHODS The chromatographic separation was performed on a capillary column of Agilent HP- 5ms( 30m × 0. 25mm,0. 25μm),and the spectrometer was operated in electron- impact ionization with an electron multiplier voltage mode. Targetions were at m /z222. 2 for patchoulic alcohol and m / z 168. 0 for pogostone. RESULTS The standard curves of patchoulic alcohol and pogostone were linear over the concentration range of 5. 040- 2016 ng·m L^- 1( r = 0. 9979) and 1. 020- 406. 0 ng·m L^- 1( r = 0. 9958),respectively.The intra- batch and inter- batch accuracies for patchoulic alcohol and pogostone at four concentrations were all in the range of ±15%. All the relative standard deviations were less than 8%. The average extraction recovery was 76. 4% and 80. 6%,respectively.CONCLUSION The method has been successfully applied to a pharmacokinetic study after the oral administration of patchoulic alcohol and pogostone in the plasma of rats.
