摘要
本试验在冷冻液中分别添加不同含量谷胱甘肽(GSH,1、3、5、7、9 mmol/L)和透明质酸(HA,300、600、900、1 200μg/m L)及联合添加GSH和HA,研究其对猪精液冷冻后活力、顶体完整率、体外受精胚胎卵裂率、超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量的影响。结果表明,添加GSH以5 mmol/L浓度效果较好,除卵裂率与未添加GSH组无显著差异外(P>0.05),活力、顶体完整率、SOD活性及MDA含量均与未添加GSH组存在统计学差异(P<0.05);添加HA以900μg/m L组效果较好,除卵裂率与未添加HA组不存在显著差异(P>0.05),其冻后活力、顶体完整率、SOD活性及MDA含量均与未添加HA组存在统计学差异(P<0.05);联合添加GSH和HA,以添加GSH 5mmol/L和HA 900μg/m L组效果较好,且其各指标与未添加GSH和HA组均存在显著差异(P<0.05)。结果说明,冷冻液中联合添加GSH 5mmol/L和HA 900μg/m L可有效改善猪精液冷冻效果。
In the experiment,we investigated the effect of supplement of different amounts of glutathione (GSH,1,3,5,7 and 9mmol/ L),hyaluronan (HA,300,600,900 and 1200μg/mL) and combination of GSH and HA prior to freezing on post-thaw sperm characteristics (motility,intact acrosome rate,cleavage rate of in vitro fertilization embryo,SOD activity and MDA content).The results suggested that 5mmol/L GSH and 900 μg/mL HA were good concentrations.There were distinct differences between 5mmol/L GSH group and free GSH group (P〈0.05) in motility,intact acrosome rate,SOD activity and MDA content except for the cleavage rate.Except that cleavage rate had no statistical difference,there was significant difference (P〉0.05) between 900μg/mL HA group and free HA groups in motility,intact acrosome rate,SOD activity and MDA content.The effect of associated addition of 5mmol/L GSH and 900μg/mL HA was better than other combination of GSH and HA groups.Our finding suggested that combination of 5 mmol/L GSH and 900 μg/mL HA could effectively improve the quality of frozen-thawed boar semen.
