摘要
以羊肚菌为材料,采用液体发酵技术,研究了碳源、氮源和培养条件对菌丝体生物量及胞外多糖产量的影响。羊肚菌液体发酵的最佳培养基为:麦芽汁1.5%,玉米粉40mg/L,黄豆粉20mg/L,酵母粉3mg/L。最优培养条件为24℃、140r/min、pH 6.0、接种量10%。在此培养条件下测定羊肚菌的生长曲线,0~48h为延滞期,48~120h为旺盛生长期;在培养144h时菌丝体生物量和胞外多糖质量浓度达到最大,分别为14.11g/L和268.9mg/L。研究结果可为大规模液体发酵生产羊肚菌胞外多糖提供一定的理论依据。
The factors affecting the biomass and yield of extracellular polysaccharides of Morchellain liquid culture were optimized.The formula of malt juice 1.5 %,corn flour 40mg/L,soybean flour20mg/L and yeast powder 3mg/L was proven to be the optimal basal media for Morchella,and the optimal culture conditions were 24 ℃,140r/min,pH 6.0and inoculum of 10%.Under the above optimal conditions,the growth curve of Morchella was determined,it was showed that the lag period of strain was 0-48 hand the logarithmic phase was 48-120 h.In addition,the maximum of biomass and yield of extracellular polysaccharides were achieved at 114 hof culture,which was 14.11g/L and268.9 mg/L,respectively.The results would provide references for large-scale production of extracellular polysaccharides by Morchella.
