复方石榴籽油对2型糖尿病小鼠血糖的影响 被引量:2

Effect of Compound Semen Granati Oil on Blood Glucose of Type 2 Diabetic Mellitus Mice

摘要 目的探讨复方石榴籽油对实验性2型糖尿病小鼠的降血糖作用。方法取健康雄性C57BL/6J小鼠,腹腔注射链脲佐菌素(STZ)120 mg·kg-1诱发高血糖模型,经过3周基础饲料喂养后,测定空腹及随机血糖。以普通基础饲料喂养的小鼠作为正常组,将造模成功的2型糖尿病小鼠,按照体质量和血糖水平随机分为模型组,二甲双胍组(130 mg·kg-1),石榴籽油组(1080 mg·kg-1),紫苏籽油组(1570 mg·kg-1),复方石榴籽油低、高剂量组(1000,3000 mg·kg-1),饲以高脂饲料,灌胃给药6个月。2,4,6个月末时,检测各组小鼠空腹血糖(FPG),并计算降糖率;2个月末时,进行葡萄糖糖耐量试验(OGTT);4个月末时,测定各组小鼠空腹胰岛素(FINS)。结果注射STZ 3周后,模型组的血糖水平高于正常组,与正常组比较,有显著性差异(P<0.05或P<0.01)。与模型组比较,石榴籽油组、紫苏籽油组及复方石榴籽油高、低剂量组小鼠的FPG、FINS水平均有不同程度的降低。复方高剂量组小鼠的FPG有显著降低(P<0.01),其降糖幅度在给药4个月时大于石榴籽油组、紫苏籽油组,且葡萄糖耐量得到显著改善(P<0.05)。结论复方石榴籽油降糖效果明显,能明显降低高胰岛素水平,且能明显改善糖耐量。 Objective To investigate the hypoglycemic activity of the compound Semen Granati oil on type 2 diabetic C57BL/6J mice. Methods Type 2 diabetic mellitus(T2DM) model was induced by intraperitoneal injection of streptozotocin(120 mg·kg^-1mice. The fasting plasma glucose and non-fasting plasma glucose levels were measured after 3 basic diet feeding for 3 weeks. Mice fed with basic diet were allocated into normal group. The successfully established experimental diabetic mice models were randomly divided into six groups according to body mass and blood glucose levels,namely model control group,metformin(130 mg·kg^-1,Fructus Perillae oil group 1570 mg·kg^-1 and low-dose compound Semen Granati oil group(the combination of Semen Granati oil and Fructus Perillae oil, in the dosage of 1000, 3000 mg·kg^-1t for the normal group were fed with high-fat diet together and were given intragastric administration of the corresponding medicine simultaneiously for six months. At the end of experimental month 2,4,6,the fasting plasma glucose was determined, and the rate of hypoglycemic was calculated. At the end of experimental month 2, the capability of glucose tolerance was observed through oral glucose tolerance test. The insulin level was determined at the end of experimental mong 4. Results After injection of STZ for 3 weeks,fasting plasma glucose level of diabetic model group was significantly higher than that of the normal group(P〈 0.05 or P〈 0.01). Compared with the model group,the fasting plasma glucose and insulin levels were decreased to various degrees in Semen Granati oil group, Fructus Perillae oil group, and compound Semen Granati oil groups. In high-dose compound Semen Granati oil group, the fasting plasma glucose was decreased significantly(P〈 0.01), glucose tolerance was improved(P〈 0.05), and the hypoglycemic activity was superior to that of Semen Granati oil group and Fructus Perillae oil group. Conclusion Compound Semen Granati oil has obvious hypoglycemic effect, and can reduce the level of insulin and improve the glucose tolerance.
