定量整体放射自显影测定D-甲基苯丙胺在小鼠脑内的分布 被引量:1

Distribution of D-methamphetamine in mice brains by quantitative whole-body autoradiography technique

摘要 目的测定D-甲基苯丙胺在小鼠脑内的分布。方法小鼠尾静脉给予2 mg/kg(10μCi 14C-甲基苯丙胺),分别在给药后5、15、30和60 min处死,应用定量整体放射自显影技术,测定D-甲基苯丙胺在脑中的分布。结果 D-甲基苯丙胺在小鼠脑内5、15和60 min时药物分布分别为(52.41±4.5)(19.50±3.88)和(18.67±1.03)n Ci/g,此外,30 min冠状切片发现药物在脑各区域是均匀分布。结论 D-甲基苯丙胺在小鼠脑内快速分布而消除较慢,且为广泛均匀分布。 Objective To determine the distribution of D-methamphetamine in mice brains. Methods Mice were sacrificed at 5, 15, 30 and 60 rain respectively, after intravenous (i.v.) bolus administration of 2 mg/kg D-methamphetamine (10 μCi 14^C-METH). Distribution of D-methamphetamine in the mice brain was investigated using quantitative whole-body autoradiography (QWBA) .Results The D-methamphetamine distribution in the mice brains were (52.41±4.5), (19.50±3.88) and (18.67±1.03) nCi/g respectively at 5, 15 and 60 min. Further more, the images of coronal section at 30 min time point were taken and measured by Multi Gauge (FUJIFILIM Co.,Ltd ), the regional distribution of the drug in the mice brain was homogenous. Conclusion D-methamphetamine is rapidly and wildly distributed in the mice brain, but clearance is slower.
