摘要
目的建立延龄草(Trillium tschonoskii Maxim.)药材质量评价的方法。方法采用3种偏诺皂苷作为指标,建立薄层色谱鉴别药材,Rp-HPLC测定含量的质量评价方法。结果 ParisⅧ(1)、偏诺皂苷元-3β-O-α-L-吡喃鼠李糖基-(1→4)-[-O-α-L-吡喃鼠李糖基-(1→2)]-O-β-D-吡喃葡萄糖苷(2)、ParisⅥ(3)在薄层色谱(三氯甲烷-甲醇-甲酸-水=6.5∶3.5∶0.5∶1)下层液中展开,斑点清晰可见、分离度好;在Rp-HPLC(乙腈-水,42∶58)中的分离度良好,分别在0.115~0.690 mg·mL^-1(r=0.999 8),0.135~0.810 mg·mL^-1(r=0.999 6),0.225~1.350 mg·mL^-1(r=0.999 9)内峰面积与进样量呈良好的线性关系,平均加样回收率为99.3%,98.3%,97.7%。结论该方法快速、准确、重现性好,可作为延龄草质量标准制定的依据。
OBJECTIVE To develop a method for the quantitative evaluation Trillium tschonoskii. METHODS Selecting three penogenin saponins as indicators,a TLC assay was developed for the identification Trillium tschonoskii,and a RP-HPLC assay was established for the content determination. RESULTS Three penogenin saponins were clearly distinguished by TLC( CHCl3-Me OHHCOOH-H2 O,6. 5∶ 3. 5∶ 0. 5∶ 1) and well separated by RP-HPLC( CH3CN-H2 O,42∶ 58). The TLC method showed clear spots,and the RP-HPLC method had good linear relationship within the ranges 0. 115- 0. 690 mg· mL^-1 for Paris Ⅷ( 1)( r = 0. 999 8),0. 135- 0. 810 mg·mL^-1 for penogenin-3 β-O-α-L-rhamnopyranosyl-( 1 →4)-[-O-α-L-rhamnopyranosyl-( 1 →2) ]-O-β-D-glucopyraboside( 2)( r = 0. 999 6),and 0. 225- 1. 350 mg· mL^-1 for Paris Ⅵ( 3)( r = 0. 999 9). The average recoveries were99. 3 %,98. 3 %,and 97. 7 %,respectively. CONCLUSION The established method is rapid and sensitive for controlling the quality T. tschonoskii.
