摘要
目的探讨炙马钱子对实验性自身免疫性重症肌无力(EAMG)模型大鼠的免疫调节机制。方法采用鼠源乙酰胆碱受体α亚基97-116肽段方法免疫Lewis大鼠建立EAMG模型,观察实验大鼠的临床表现,并将建模成功的EAMG大鼠随机分为模型组、强的松组及炙马钱子低、中、高剂量组,每组8只。采用酶联免疫吸附试验(ELISA)法检测各组大鼠血清乙酰胆碱受体抗体(ACh Rab)和白介素6(IL-6)含量比较各组之间差异。结果模型组大鼠血清ACh Rab和IL-6含量较正常组显著升高(P均<0.01)。炙马钱子高剂量组ACh Rab含量较中、低剂量组下降更显著(P均<0.01),与强的松组比较,差异无统计学意义(P>0.05);炙马钱子中、高剂量组IL-6含量均低于强的松组(P均<0.01);炙马钱子中、高剂量组间IL-6含量差异无统计学意义(P>0.05)。结论炙马钱子能降低EAMG模型大鼠血清ACh Rab和IL-6含量,抑制T、B细胞活化,减轻对突触后膜Ach R的损害,缓解EAMG病情,一定剂量范围内的炙马钱子在免疫调节方面表现优于强的松。
Objective To investigate the immune regulation mechanism of nux vomica on experimental autoimmune myasthenia gravis (EAMG) rats. Methods Lewis rats were immunized with torpedo-derived 97-116 peptide of the AchRα-subunit in CFA. After the second immunization, the rats" weights and symptoms were observed to judge the success of the EAMG model. Rats with EAMG were divided into model group, prednisone group, nux vomica groups (low-, medium-, and high-dosage), 8 rats in each group, and were given the corresponding treatment, respectively. The levels of AchRab and IL-6 were determined by using enzyme linked immunosorbent assay (ELISA). Results The level of AChRab and IL-6 were increased in model group (all P〈0.01). The level of AChRab in high-dose nux vomica group was lower than that in medium- and low-dosage nux vomica groups (P〈 0.01), but was not different with that in prednisone group (P〉0.05). The level of IL-6 in medium- and high- dosage nux vomica groups was significantly lower than that in prednisone group(all P〈0.01 ), but there was no sig- nificant difference between high- and medium-dosage nux vomiea groups (P〉0.05). Conclusion Nux vomica can reduce the level of AChRab and IL-6 in EAMG model, inhibit the activation of T cells and B cells, so that it can reduce the damage of postsynaptic membrane AchR, and alleviate the development of EAMG. The effect of nux vomica is better than prednisone in improving immune adjustment in a certain dosage range.
