摘要
目的 探讨氯沙坦和卡托普利联合应用对自发性高血压大鼠 (SHR)肾结构与功能的保护作用。方法 将自发性高血压大鼠 30只随机分为对照组和 3个治疗组 ,治疗组分别以氯沙坦 (30mg·kg- 1 ·d- 1 )、卡托普利 (1 0 0mg·kg- 1 ·d- 1 )、卡托普利50mg·kg- 1 ·d- 1 +氯沙坦 1 5mg·kg- 1 ·d- 1 灌胃 1 5周 ,观察大鼠的血压、尿蛋白 ,肾组织中血管紧张素Ⅱ (AngⅡ )、转化生长因子 β1 (TGFβ1 )以及肾组织病理改变。结果 氯沙坦和卡托普利治疗组的尿蛋白显著减少 ,肾内Ⅳ型胶原沉积和TGFβ1 减少 ,肾小动脉结构正常 ,且氯沙坦 +卡托普利治疗作用更显著。结论 氯沙坦合用卡托普利后具有协同作用 ,可以延缓肾脏损害。
OBJECTIVE: To observe the effects of losartan and captopril on renal structure and function in spontaneously hypertensive rats(SHR). METHODS: Thirty SHRs were gorged with losartan (30 mg·kg-1·d-1), captopril (100 mg·kg-1·d-1), losartan (15 mg·kg-1·d-1) + captopril (50 mg·kg-1·d-1) or water respectively for 15 weeks in the treating and control groups. The blood pressure and proteinuria were assessed, the levels of angiotensin II (Ang II) and transforming growth factor(TGF)β1 were detected, and renal tissues were examined routinely by optical and electron microscopy respectively. RESULTS: The excretion of proteinuria was significantly decreased in the treating groups compared with the control group. There was a decrease TGFβ1 and collagen IV deposition in the treatment group, and further lower in losartan + captopril group. Losartan and captopril attenuated the injuries of small arteries. CONCLUSION: The combination of losartan with captopril may attenuate renal damage, they had synergistic action when used together.
