摘要 【目的】探讨猪GHR基因两个SNPs位点多态性与其生长性状的相关性,为保护广西巴马小型猪的遗传资源及推进猪分子标记辅助育种技术的发展奠定基础。【方法】采用PCR-RFLP和错配PCR-RFLP(CRS-PCR-RFLP)检测巴马小型猪×长白猪杂交F2代(简称巴×长F2代)资源家系中猪GHR基因G1248A和A1801G两个SNPs位点的多态性,并对其相关生长性状进行关联分析。【结果】猪GHR基因G1248A和A1801G两个SNPs位点在巴×长F2代资源家系中均表现出AA、AG和GG 3种基因型。G1248A位点对巴×长F2代生长性状的影响表现为:除2月龄GG基因型个体的胸围尺寸显著大于AA、AG基因型个体(P<0.05,下同),4月龄GG基因型个体的体重显著高于AA、AG基因型个体外,其他月龄的相关生长性状在不同基因型个体间差异均不显著(P>0.05,下同)。A1801G位点对巴×长F2代生长性状的影响表现为:除1和6月龄GG基因型个体的体高显著低于AA、AG基因型个体,3和6月龄GG基因型个体的胸围尺寸显著大于AA、AG基因型个体外,其他月龄的相关生长性状在不同基因型个体间差异也不显著。【结论】猪GHR基因G1248A位点的G等位基因对个体体重、胸围有正选择作用,而A1801G位点的G等位基因对个体体高有负选择作用,对个体胸围有正选择作用,均可作为猪分子标记辅助育种的遗传选择标记。 【Objective 】The correlation of 2 SNPs sites polymorphism in GHR gene and growth traits of pig were investigated in order to lay the foundation for protecting Guangxi Bama mini-pig genetic resources and advancing pig molecular marker assisted breeding technology. 【 Method 】 The polymorphism of 2 SNPs sites viz. GHR- G1248 A and A1801 G in hybrids F2 family pigs of Guangxi Bama mini-pig × Landrace were detected through PCR-RFLP and CRS-PCR-RFLP. The correlation between 2 SNPs sites and pig 's corresponding growth traits was analyzed. 【Result 】In hybrids F2 family of Guangxi Bama mini-pig × Landrace, 2 SNPs sites of pig GHR viz. G1248 A and A1801 G sites were both expressed as three genotypes, AA, AG and GG, respectively. As to the influences of G1248 A site on growth traits, it showed that the chest circumferences of 2-month old pigs for genotype GG were significant larger than that of genotype AA and AG, the body weights of 4-month old pigs for GG genotype were significant heavier than that of genotype AG and AA, while the growth traits differences of other months old pigs were found not significant among different genotypes ' individuals( P〉0. 05, the same below). As for the effects of A1801 G site on growth traits, it found that the body heights of 1- and 6-month old pigs for GG genotype were significant higher than that of genotype AG and AA, the chest circumferences of 3- and 6-month old pigs for genotype GG were significant larger than that of genotype AA and AG, while the growth trait differences of other months old pigs were found not significant among different genotypes ' individuals(P〉0.05,the same below). 【Conclusion 】G allele of pig GHR/G1248 A site displays positive selection effects on body weight and chest circumference, while G allele of pig GHR/A1801 G site displays negative selection effect on body height and positive selection on chest circumference, therefore those sites can be selected as assistant-selected genetic markers for pig breeding.
