摘要
为合理选择实验优化设计方法并准确快捷地获得最佳工艺条件,将单因素、正交试验和响应面法分别用于荞麦中芦丁提取工艺的优化,分析了提取时间、乙醇浓度、提取温度和液固比对芦丁提取效率的影响。结果表明,单因素和正交试验确定的最佳工艺为:提取时间120 min、乙醇浓度100%、提取温度90℃、液固比30∶1;而响应面法确定的最佳工艺为:提取时间114 min、乙醇浓度96%、提取温度86℃、液固比32∶1,三种方法所得结果基本一致。最终,以上述所得最优工艺条件进行试验验证,结果表明按照响应面法确定的最优工艺条件所得的提取率高于单因素/正交试验6.91%。因此认为响应面法确定的工艺条件较好,荞麦中芦丁的提取率可达0.3452 mg/g。
For the reasonable selection of experimental design method and accurately obtain the optimal process conditions, Rutin extraction process from Buckwheat were investigated by single factor analysis, orthogonal experiment and response surface method. The effects of extraction time, ethanol concentration, temperature and the ratio of liquid to materials on the extraction rate were discussed. The optimal process obtained by single factor analysis and orthogonal design is extraction time 120 min, ethanol concentration 100 %, temperature 90 ℃, and the ratio of liquid to materials 30 : 1; while that by response surface method is extraction time 114 rain, ethanol concentration 96 %, temperature 86℃, and the ratio of liquid to materials 32 : 1. The results show that three methods obtained the same conclusion. Verification testing results indicate that the extraction rate under the optimal process of response surface method is 6.91% higher than that of single factor analysis and orthogonal design. Therefore, the optimal process was decided according to response surface method, under which the extraction rate can reach 0.3452 mg/g.
