2010—2012年浙江省余姚市手足口病的病原构成及Cox A16 VP1基因特征分析 被引量:1

摘要 目的了解浙江省余姚市引起手足口病的病原构成,研究Cox A16基因特征及变异情况。方法收集手足口病患者标本,用实时荧光定量聚合酶链反应(real-time PCR)检测并分型,每年选择几个样品进行Cox A16 VP1基因的扩增和核苷酸序列测定,测序结果利用DNAstar和Blast软件进行比对分析。结果 2010—2012年从1651份标本检出柯萨奇病毒A组16型(Cox A16)25.86%,肠道病毒71型(EV71)39.85%,其他肠道病毒19.38%;Cox A16 VP1蛋白氨基酸序列相似性为99.3%-100%,7株Cox A16基因分型属于B1基因亚型。结论 2010—2012年余姚市手足口病流行的主要病原体为Cox A16和EV71,流行的Cox A16属于B1基因亚型,并具有高度相似性。 Objective To understand the constituent of pathogens causing hand foot and mouth disease( HFMD) and the genetic characteristics of Coxsackie virus A 16( Cox A16). Methods The stool samples collected from HFM D patients w ere detected w ith real time fluorescence polymerase chain reaction. Several stool samples w ere selected every year to conduct the VP1 gene amplification and nucleotide sequencing of Cox A16,and the sequencing results w ere compared and analyzed w ith softw are DNAstar and Blast. Results From 2010 to 2012,a total of 1651 stool samples w ere detected,in w hich 25. 86% w ere Cox A16 positive,39. 85% w ere EV71 positive and 19. 38% w ere other enterovirus positive. The VP1 protein amino acid sequence of the Cox A16 strains detected shared 99. 3% to 100% similarity. The 7 Cox A16 strains belonged to genotype B1. Conclusion Cox A16 and EV71 w ere the predominant pathogens causing HFM D in Yuyao during 2010-2012,and the circulating Cox A16 belonged to genotype B1 w ith high similarity.
