小陷胸汤合丹参饮含药血清对Ang-Ⅱ损伤人脐静脉内皮细胞ICAM-1表达的影响 被引量:3

摘要 目的从影响淋巴细胞与血管内皮细胞粘附的角度探讨小陷胸汤联合丹参饮抗动脉硬化作用的机制。方法采用Ang-Ⅱ损伤体外培养的人脐静脉内皮细胞(HUVEC)作为病理模型,研究小陷胸汤合丹参饮对淋巴细胞与血管内皮细胞粘附的影响及其分子机制。结果用Ang-Ⅱ损伤HUVEC后,细胞ICAM-1mRNA表达、ICAM-1蛋白含量以及淋巴细胞粘附率显著升高,而不同浓度的小陷胸汤合丹参饮含药血清则能降低ICAM-1mRNA表达、ICAM-1蛋白含量,减少HUVEC与淋巴细胞粘附,且基本呈浓度相关。结论通过降低细胞表面粘附分子的表达而减少淋巴细胞与内皮细胞的粘附,可能是小陷胸汤合丹参饮抗AS作用的机制之一。 Objective To investigate the effects of Xiaoxianxiong decotion and Danshen decoction (XAD) on anti - arteriosclerosis effect mechanisms by lymphocytes and endothelial cell adhesion. Methods Based on the pathological model of human umbilical vein endothelial cell ( HUVEC), the effects of XAD on lymphocyte - endothelium adhesion and the molecular mechanism were revealed. Results After injury HUVEC with Ang- Ⅱ , the expressing of the ICAM -1mRNA, ICAM -1 protein levels and lymphocyte adhesion rate increased significantly, and different concentrations XAD containing serum was able to reduce the expression of ICAM - 1 mRNA , ICAM - 1 protein content decreased HUVEC adherence for lymphocyte, and basically in a dose - related. Conclusion One of the mechanisms of anti - AS of XAD is that it decreased lymphocyte - endothelial adhesion by reducing the expression of cell surface adhesion molecules.
