摘要
目的:研究慢性HBV携带者外周血中辅助性T淋巴细胞1(Th1)/辅助性T淋巴细胞2(Th2)相关细胞因子白细胞介素2(IL-2)、白细胞介素4(IL-4)及γ干扰素(γ-IFN)随患者体内病毒含量变化时的变化及其意义。方法:通过荧光定量方法检测86名慢性HBV携带者体内的病毒载量,而后根据病毒载量值将患者分为HBV DNA阴性组、HBV DNA低载量组、HBV DNA高载量组;通过ELISA方法检测86名慢性HBV携带者及39名正常人血清中IL-2、IL-4及γ-IFN的含量。结果:各患者组外周血清中IL-2的含量比正常组增高,且HBV DNA低载量组、HBV DNA高载量组与正常组比较,差异有统计学意义(F=3.055,P=0.031);各患者组外周血清中IL-4的含量比正常组增高,HBV DNA高载量组与正常组比较,差异有统计学意义(F=2.486,P=0.064);各患者组外周血清中γ-IFN的含量与正常组相比,差异无统计学意义(F=0.063,P=0.976)。结论:慢性HBV携带者体内存在Th1/Th2平衡紊乱,且可能Th1存在免疫功能障碍是形成的机制之一。
Objective: To investigate the changes of Thl/Th2 related cytokines IL-2, IL-4 andγ-IFN in peripheral blood of patients with chronic HBV carriers. Methods: HBV DNA loads were detected by fluorescent quantitative PCR. in 86 chronic HBV carriers, then patients were divided into three groups: HBV-negative group, low HBV DNA loads group and high HBV DNA loads group. The levels of IL-2, IL-4 and γ-IFN in peripheral blood of 86 chronic HBV carriers and 39 normal controls were analyzed by ELISA. Results: The levels of IL-2 in chronic HBV carriers increased compared with normal controls group, low HBV DNA loads group and high HBV DNA loads group significantly increased compared with normal controls group , ( F = 3. 055, P = 0. 031 ) ; The levels of IL-4 in chronic HBV carriers increased compared with normal controls group, high HBV DNA loads group significantly increased compared with normal controls group ( F = 2. 486, P = 0. 064) ; The levels of γ-IFN were no significant differences among three groups ( F = 0. 063, P = 0. 976 ) . Conclusion : There have dysfunction in T helper cell (Th1/Th2) of patients with chronic HBV carriers, and it may be one of the mechanisms of immune dysfunction in Thl cells of patients with chronic HBV carriers.
