摘要
目的观察、了解低渗造影剂碘比乐诱导的造影剂急性肾损伤(AKI)不同时间段大鼠血半胱氨酸蛋白酶抑制蛋白C(Cys C)、尿肝脏型脂肪酸结合蛋白(L-FABP)表达变化及其意义。方法将60只SD大鼠,随机分为造影剂急性肾损伤(AKI)组(AKI组,n=30)和非AKI(非AKI组,n=30),每组分为6个亚组,包括2、4、6、12、24、48 h组,观察各亚组血Cys C、尿L-FABP不同时间段的表达变化。结果与非AKI组相比较,AKI组血Cys C的表达:变化造模后12、24、48 h大鼠Cys C水平均显著升高,且两两比较差别有统计学意义(P均<0.05)。尿L-FABP水平在第8、12、24、48 h非AKI与AKI组相比较差别有统计学意义(P<0.05)。Scr的相关性分析显示,不同AKI组尿L-FABP、Cys C呈正相关(r分别为0.900、0.954,P<0.05);Cys C的相关性分析显示,不同AKI组尿L-FABP、Scr呈正相关(r分别为0.937、0.953,P<0.05);AKI组尿L-FABP的相关性分析显示,不同AKI组尿L-FABP、Scr呈正相关(r分别为0.937、0.953,P<0.05)。结论尿L-FABP为急性肾损伤早期敏感的生物标记物。
Objective To observe low permeability Iopamidol, a kind of iodine contrast agent, induced acute kidney injury(AKI) and investigate the changes and the significance of serum cystatin C (CysC)and liver type fatty acid - binding protein( L - FABP) levels at different time in rats. Methods 60 SD rats were randomly divided into the contrast induced acute kidney injury (AKI) group ( group AKI, n = 30) and non AKI group ( non AKI group, n =30) ; furthermore, each group was divided into 6 sub -groups (including 2h,4h,6h, 12h,24h,48h group). Blood and urine L - FABP at different time in rats were detected. Results Compared with the non AKI group, the CysC level in AKI group was increased at 12 h, 24 h and 48 h time point and showed significant difference (P 〈0.05). The levels of urinary L-FABP at 8 h,12 h,24 h and 48 h time points were increased in AKI group compared with the non AKI group, the difference was significant (P 〈 0.05 ). Scr correlation analysis showed posi- tive relation in different AKI sub - group, r coefficient of urinary L - FABP and CysC was 0. 900 and 0. 954, respec-tively,P 〈0.05 ;CysC correlation analysis showed positive relation as well, r coefficient of urinary L- FABP and CysC was 0. 937 and 0. 953, respectively, P 〈 0.05 ; L - FABP correlation analysis also showed positive relation, r coefficient of urinary L - FABP and CysC was 0. 937 and 0. 953, respectively, P 〈 0.05. Conclusions Urinary L - FABP is as a sensitive bio - marker for early acute kidney injury.
