摘要
目的了解缓激肽受体B2在肾脏相关细胞中的蛋白和核酸表达水平,以及在体外高糖环境下该受体对细胞增殖能力的影响。方法以Western Blot、RT-PCR技术分别对人体肾脏相关细胞中缓激肽受体B2进行检测,分别敲低肾小球系膜细胞和肾小球毛细血管内皮细胞中的缓激肽受体B2,用MTT方法检测两种细胞在高糖环境下(葡萄糖终浓度为10mmol/L)的增殖能力。结果肾小球系膜细胞、肾小球毛细血管内皮细胞中均可检测出缓激肽受体B2的表达。同时发现高糖环境下,敲低缓激肽受体B2水平,可以促进细胞的增殖能力。结论缓激肽受体B2在肾小球系膜细胞、肾小球毛细血管内皮细胞中有表达,在体外高糖环境下该受体对上述细胞增殖能力存在影响。
Objective To clarify the expression level of bradykinin B2 receptor in protein and nucleic acid of the kidney associated cells and the effect of this receptor on cell proliferation under high sugar environment in vitro. Methods Bradykinin B2 receptor in human kidney associated ceils was detected by Western Blot and RT - PCR technology, and Knocking down the bradykinin B2 receptor in the glomerular mesangial cells and glomerular capil- lary endothelial cells, respectively. The ability of proliferation concerning both kinds of cell mentioned above under a high sugar(the glucose terminal concentration: 10 mmol/L)environment was detected by MTr method. Results The bradykinin B2 receptor expression could be detected among glomerular mesangial ceils and glomerular capillary endothelial cells and the cell proliferation could be promoted in the condition of knocking down the level of bradyki- nin B2 receptor under a high sugar environment. Conclusion Bradykinin B2 receptor can be expressed in the glo- merular mesangial cells and glomerular capillary endothelial ceils and this receptor may affect the cell proliferation ability under a high sugar environment in vitro as well.
