当归多糖对脑梗死大鼠血脑屏障通透性影响及MRI评价 被引量:4

摘要 目的研究当归多糖对脑梗死大鼠血脑屏障(BBB)通透性影响及MRI评价。方法采用线栓法制作大鼠脑梗死模型,采用伊文斯蓝(EB)法检测BBB的通透性,Western blot测定紧密连接蛋白Claudin-5、ZO-1的表达,MRI法检测梗死体积。结果大鼠脑梗死24 h后,当归多糖组缺血侧脑组织EB含量显著低于模型组(P<0.01);当归多糖治疗组缺血侧脑组织Claudin-5及ZO-1表达显著高于模型组(P<0.01);且当归多糖组梗死面积显著低于模型组(P<0.01)。结论当归多糖能降低脑梗死大鼠BBB通透性,与Claudin-5及ZO-1表达上调密切相关,从而减少脑梗死体积。 Objective To investigate the effects of angelica polysaccharide on blood- brain barrier( BBB) permeability in rat cerebral infarction and MRI evaluation. Methods Rats were made into Rat cerebral infarction model was established with suture method. BBB permeability was measured by Evans Blue( EB). The expressions of claudin- 5 and ZO- 1 were measured by western blotting assay. The volume of cerebral infarction was measured by MRI. Results Twenty- four h after cerebral infarction,the EB content was significantly decreased,the expressions of Claudin- 5 and ZO- 1 were significantly increased and the volume of cerebral infarction was significantly decreased in angelica polysaccharide group( P〈0. 01) compared with the model group. Conclusion Angelica polysaccharide could decrease the BBB permeability and increase Claudin- 5 and ZO- 1 expressions and decrease the volume of cerebral infarction in rat cerebral infarction.
