摘要
目的:建立大鼠Walker-256乳腺癌胫骨癌痛模型,予以通络散结凝胶外用并探讨其镇痛疗效。方法:取雌性Wistar大鼠40只,右后肢胫骨注射Walker-256乳腺癌细胞建立骨癌痛模型。将大鼠随机分成5组:空白凝胶组、扶他林乳剂组、通络散结凝胶Ⅰ组、通络散结凝胶Ⅱ组、通络散结凝胶Ⅲ组。每组8只,各组大鼠自造模后第1天开始给药,1 d/1次,连用21天。同时动态观察大鼠体重以及机械痛觉缩足阈值、热痛觉缩足潜伏期和自发痛评分。结果:1造模后第5天,空白凝胶组大鼠体重下降,中药组治疗后体重逐渐增加,其中通络散结凝胶Ⅲ组体重增加最为明显(P〈0.001)。2与空白凝胶组比较,通络散结凝胶从第17天开始,能够显著提高大鼠机械痛阈值,以通络散结凝胶Ⅲ组为最佳。3造模后第18~20天,与空白凝胶组比较,通络散结凝胶能够提高大鼠热痛觉缩足阈值。4各组大鼠在造模5天后大鼠逐步有跛行现象,造模后第21天,与空白凝胶组比较,通络散结凝胶能够降低大鼠自发痛评分,其中通络散结凝胶Ⅲ组效果较好。结论:通络散结凝胶对骨癌痛具有一定镇痛效果,且药物浓度越高,止痛效果越佳。
Objective:Bone cancer pain rat model was made by injecting Walker- 256 breast carcinoma cells to the right tibia of rats. Tongluo Sanjie gel (TLSJ) was externally used to the right tibia of rats and the pain behavior was measured to detect the effect of TLSJ gel on controlling bone cancer pain. Methods :The right hind tibia of rats were injected Walker -256 tunmr cells to build bone cancer pain model. 40 female SD rats were randomly divided into 5 groups:blank gel group,votalin group,TLSJ gel I group,TLSJ gel Ⅱ group,TLSJ gel Ⅲ group, n = 8. Each group was administrated on 1 ~ 21 days after surgery. Spontaneous pain, mechanical threshold, paw withdrawal latency (PWL) were measured, and the security of TLSJ gel was evaluated. Results:1. On day 5 after surgery ,the weight of blank gel group showed decrease while the TLSJ gel group's increased and the TISJ gel m group's increased obviously (P 〈0. 001 ). 2. Compared to blank gel group,on day 17 -21 after surgery,TLSJ gel can increase the mechanical allodynia,and TLSJ gel Ⅲ was the best. 3. Compared to blank gel group, on day 18 ~ 20 after surgery, TLSJ gel can increase the PWL,and TLSJ gel Ⅲ was the best. 4. On day 5 -21 after surgery, gradually claudication of each group rats was observed. On day 21, compared to blank gel group, TLSJ gel can decrease the spontaneous pain, and TLSJ gel Ⅲ was the best. Conclusion:The research implies that TLSJ gel can control bone cancer pain and the effect of controlling pain was gradually increased with the increased concentration of atorvastatin.
