摘要
目的:观察糖脂清颗粒对2型糖尿病大鼠心肌组织Bax、Bcl-2表达的影响,并探讨其干预心肌细胞凋亡的机制。方法:雄性SD大鼠,采用高糖高脂饲料喂养8周后,一次性小剂量(30 mg·kg-1)2%链脲佐菌素(STZ)溶液腹腔注射造模,3 d后尾尖采血,测随机血糖≥16.7 mmol·L-1者确定为2型糖尿病大鼠模型,8周后,用免疫组织化学法检测各组心肌组织Bcl-2、Bax的表达。结果:糖脂清颗粒各治疗组大鼠心肌组织Bcl-2表达明显上升,与模型组比较,差异有统计学意义(P<0.05);各治疗组大鼠心肌组织Bax/Bcl-2明显下降,与模型组比较,差异有统计学意义(P<0.05)。结论:糖脂清颗粒具有抗糖尿病大鼠心肌细胞凋亡的作用,其可能机制为升高Bcl-2的表达,从而降低Bax/Bcl-2的比值,抑制心肌细胞凋亡,延缓糖尿病心肌病的进展。
Objective : To observe the effect of Tangzhiqing Granule on the expression of Bax and Bcl-2 in myocardium of type 2 diabetic rats, and to explore the intervention of myocardial cell apoptosis mechanism. Methods : Male SD rats with high fat and high sugar diet for 8 weeks, disposable small dose ( 30 mg·kg^-1 ) 2% streptozotocin (STZ) solution intraperitoneal injection for molding, after 3 days, collection the tail tip blood, measuring random blood glucose ≥16.7 mmol·L^-1 to identify type 2 diabetes rat model, and 8 weeks later, the expression of immunohistoehemistry was used to detect Bcl-2 and Bax in myocardial tissue in each group. Results : The expressions of BcL-2 in myocardium of rats in Tangzhiqing Granule each treatment group were increased significantly, compared with the model group, the difference was statistically significant( P 〈 0.05 ) ;and Bax/Bcl-2 were decreased significantly, compared with the model group, the difference was statistically significant( P 〈 0.05 ). Conclusion:Tangzbiqing Granule has the effect of anti diabetic rats myocardial cell apoptosis, and the possible mechanism is increasing the expression of Be1-2, thereby reducing the ratio of Bax/Bcl-2, inhibiting the apoptosis of myocardial cells, and delaying the progression of diabetic cardiomyopathy.
