电针足三里对术后大鼠腹腔粘连组织血管内皮生长因子表达的影响 被引量:1

Effect of Electro-Acunpunture at Zusanli Point on Expression of VEGF in Postoperative Intra-Abdominal Adhesions Rats

摘要 目的:观察电针足三里对术后大鼠腹腔粘连组织血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响,探讨电针足三里发挥抗炎作用的机制。方法:雄性Wistar大鼠40只,随机分为:A组(模型组)、B组(模型+电针足三里组)、C组(模型+电针非穴组)、D组(造模后注射α-银环蛇毒素组)和E组(造模后注射α-银环蛇毒素+电针足三里组),每组8只。采用Chiang方法制作大鼠腹腔粘连模型,B组和E组术后给予持续电针刺激双侧足三里穴1 h,连续3 d;C组采用相同频率和时间刺激非经非穴,D组和E组术毕腹腔注射α银环蛇毒素(α-BGT)。各组大鼠于术后第10天处死,剪取磨损盲肠组织,行VEGF免疫组织化学染色,观察镜下组织VEGF表达情况。结果:术后第10天,各组大鼠剖腹可见均发生腹腔粘连,镜下可见VEGF表达均有不同程度增加;B组粘连组织VEGF表达明显低于其余各组(P<0.01)。与A组比较,C组、D组及E组VEGF表达无明显降低或升高,差异无统计学意义(P>0.05)。结论:电针足三里可有效降低术后腹腔粘连组织VEGF过度表达;阻断胆碱能受体α7亚基后施行电针足三里,组织VEGF表达无明显降低,其发挥抗炎机制与胆碱能抗炎通路有关。 Objective: To observe the effect of electro-acupuncture( EA) at Zusanli points on vascular endothelial growth factor( VEGF)of postoperative intra-abdominal adhesions,and to explore its anti-inflammatory pathway. Methods: Forty male Wistar rats were divided into 5 groups( each = 8) : Group A( abdominal adhesions model),Group B( abdominal adhesions plus EA),Group C( shame acupoint control),Group D( abdominal adhesions plus α-bungarotoxin) and Group E( abdominal adhesions plus EA after α- bungarotoxin). Animal models of abdominal adhesion were produced by Chiang's path. Bilateral Zusanli points and shame acupoints were electro-acupunctured with constant voltage for 1 hour at rats' waking. The α-BGT( 1 μg·kg^-1) was injected into abdominal cavity after the surgery. All the rats were sacrificed on the 10 th day,the the VEGF in tissues were evaluated. Results: The rats were sacrificed on the tenth day,the damaged cecum of abdominal adhesion groups developed obvious edema,and respectively adhered with other tissues; Compared withgroup B,the expression of VEGF was higher( P〈0. 01); Compared with group A,group C,group D and group E have no decrease VEGF expression or higher,there was no statistically significant difference( P〉0. 05). Conclusion: EA at Zusanli points significantly lowered the expression of VEGF after abdominal adhesion challenge; The activation of cholinergic anti-inflammatory pathway might be one of the mechanisms of EA at Zusanli points exerting the anti-inflammatory.
