短穗鱼尾葵果实原花青素粗提物的树脂纯化

摘要 短穗鱼尾葵果实产量大、原花青素含量高,研究短穗鱼尾葵原花青素的树脂纯化技术,为其开发利用提供试验依据.通过5种大孔吸附树脂供试,筛选出的DS401在25℃对短穗鱼尾葵原花青素的饱和吸附量为45.42mg/g、解吸率为89.91%.DS401树脂在粗提物溶液p H 5、温度45℃、吸附时间4.5 h对原花青素吸附的效果好,以80%乙醇为解吸液、解吸液p H=8、温度45℃进行DS401树脂吸附原花青素的解吸效果最佳.以体积分数80%的乙醇溶液、0.75BV/h流速动态洗脱,在2.5 BV的洗脱峰中原花青回收率为93.90%,洗脱时间4.5 h. Caryota mitis bears a lot of fruit annually and proanthocyanidins is full in the fruit yield. The research on the purification technology of the crude procyanidins extract with resin is to provide the experimental basis for the development and utilization. Proeyanidins adsorption capacity of the DS401 rmaeroporous resins was 45.42mg g, the desorption rate of 89.91% with five kinds of macroporous resins for the tested at 25℃. The optimal conditions of DS401 adsorption of the proeyanidins are pH 5, 45℃ for 4.5h. The optimal desorption conditions of the proeya- nidins on DS401 are with 80% ethanol as desorption agent, pH 8.0 at 45℃. The dynamic desorption proeyanidin was using 80% ethanol solution as desorption agent, flow rate 0.75BV/h and pH 5.0. The proeyanidin recovery was 93.90% in the 2.5 BV times main peaks, and desorpted for 4.5h.
