化疗前后乳腺癌患者血清中IL-6、IL-8及TNF-α的变化及意义 被引量:12

Changes and significance of IL- 6,IL- 8 and TNF- α in serum of breast cancer patients before and after chemotherapy

摘要 目的:探讨化疗前后乳腺癌患者血清中IL-6、IL-8及TNF-α的变化及意义。方法:选择住院手术乳腺癌术后1个月、化疗1个疗程(21天)患者50例,化疗前、后均静脉采血,同期选取体检健康女性30例作为对照组亦静脉采血,均留取血清待测。采用ELISA酶联免疫法检测IL-6、IL-8及TNF-α的蛋白水平,采用流式细胞免疫学法检测IL-6、IL-8及TNF-α的细胞阳性百分率。结果:与对照组比较,化疗前乳腺癌组患者血清中IL-6、IL-8、TNF-α的蛋白水平及细胞阳性百分率均升高,差异有统计学意义(P<0.01);化疗后患者血清中IL-6、IL-8、TNF-α的蛋白水平及细胞阳性百分率均略升高,差异无统计学意义(P>0.05);化疗前乳腺癌组患者血清中IL-6、IL-8、TNF-α的蛋白水平及细胞阳性百分率明显高于化疗后患者,差异有统计学意义(P<0.01)。结论:化疗能够通过降低乳腺癌患者血清中IL-6、IL-8、TNF-α的水平抑制肿瘤的浸润和转移,这为乳腺癌的临床治疗提供新的靶点。 Objective: To explore the changes and significance of interleukin -6 (IL -6), interleukin -8 (IL -8) and tumor necrosis factor - α ( TNF - α) in serum of breast cancer patients before and after chemotherapy. Methods: Fifty breast cancer patients were selected from this hospital at one month after surgery and after one course of chemotherapy, venous blood samples were obtained before and after chemotherapy; thirty healthy women after physical examination were selected during the same period, venous blood samples were ob- tained. ELISA was used to detect IL - 6, IL - 8 and TNF - α levels; flow cytometric immunological method was used to detect the percenta- ges of positive cells expressing IL - 6, IL - 8 and TNF - at. Results: Compared with control group, the levels of IL - 6, IL - 8 and TNF - at in serum and the percentages of positive ceils expressing IL - 6, IL - 8 and TNF - at in breast cancer group before chemotherapy increased significantly, there were statistically significant differences ( P 〈 0. 01 ) ; after chemotherapy, the levels of IL - 6, IL - 8 and TNF - at in ser- um and the percentages of positive cells expressing IL - 6, IL - 8 and TNF - at in breast cancer group increased slightly, there was no statis- tically significant difference (P 〉 0.05 ) ; in breast cancer group, the levels of IL - 6, IL - 8 and TNF - α in serum and the percentages of positive cells expressing IL - 6, IL - 8 and TNF - α before chemotherapy were statistically significantly higher than those after chemotherapy (P 〈 0. 01 ) . Conclusion: Chemotherapy can inhibit invasion and metastasis of breast cancer by reducing the levels of IL- 6, IL -8 and TNF - α in serum, which provides a new target for treatment of breast cancer.
