摘要
目的研究云南重楼乙醇提取物的化学成分及其抑制鼻咽癌细胞的活性。方法采用反复硅胶柱层析和Sephadex LH-20等色谱方法对云南重楼的大孔树脂60%(体积分数)乙醇洗脱部位进行分离、纯化,通过理化常数和波谱分析鉴定化合物结构;采用MTT比色法进行体外抑制鼻咽癌细胞(CNE)活性测试。结果从云南重楼的大孔树脂60%(体积分数)乙醇洗脱部位分离鉴定出2个脂肪酸类化合物,分别为(8R*,9R*,10S*,6Z)-trihydroxyoctadec-6-enoic acid(1)和methyl(9S,10R,11S)-trihydroxy-12(Z)-octadecenoate(2);活性测试结果显示,化合物2具有一定的抑制鼻咽癌细胞活性,其IC50为20.77μmol·L-1。结论 2个脂肪酸均为首次从该属植物中分离得到。
Objective To study on the chemical constituents of the alcoholic extract of Paris polyphylla var.yunnanensis and their antiproliferative activities on nasopharyngeal carcinoma epithelia cell( CNE).Methods The constituents were isolated and purified by repeated column chromatographies including silica gel and sephadex LH-20. Their structures were elucidated by their physicochemical properties and spectral data. The identified compounds were screened for their antiproliferative activities on CNE by using the 3-( 4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide( MTT) assay. Results Two hydroxy fatty acids were obtained from the 60% ethanol eluting fraction of macroporous resin,( 8R*,9R*,10S*,6Z)-trihydroxyoctadec-6-enoic acid( 1) and methyl( 9S,10 R,11S)-trihydroxy-12( Z)-octadecenoate( 2). The antiproliferative test results indicated that compound 2 showed weak antiproliferative effect on CNE cells with IC50 value of 20.77 μmol·L-1. Conclusion Compounds 1-2 were isolated from the genus Paris for the first time.
