流率及唾液淀粉酶活性用于评价大鼠酸刺激前后唾液样本采集方法的探讨 被引量:2

摘要 目的了解柠檬酸刺激对大鼠唾液淀粉酶(s AA)活性和唾液流率的影响,探讨味觉刺激下的唾液标本采集方法。方法随机选取正常雌性SD大鼠30只,先用微量取样器抽取刺激前的全唾液,然后用0.4 mol/L 0.50 cm×0.50 cm的柠檬酸滤纸每隔2.5 min刺激大鼠舌尖30 s,5 min采集的唾液混合为一管,连续采集15 min,共3管唾液样本,记录每管唾液采集量,用Bernfeld法测定s AA活性,计算唾液流率、酸刺激前后流率和s AA活性比值,检测流率和s AA活性的相关性。结果大鼠酸刺激后0~5 min、6~10 min、11~15 min 3个时间段的s AA活性和唾液流率以及其比值都显著增加(P〈0.05),但这些指标在刺激后的3个时间段之间差异无统计学意义(P〉0.05)。刺激前唾液流率与s AA呈显著正相关(P〈0.05),刺激后呈显著负相关(P〈0.05)。结论柠檬酸刺激能显著增加大鼠唾液流率和s AA活性。以0.4 mol/L 0.50 cm×0.50 cm柠檬酸滤纸,每隔2.5 min刺激大鼠舌尖30 s,采集酸刺激后全唾液的方法,简便易行,适用于大鼠酸刺激后唾液标本的采集。 Objective To evaluate the effect of citric acid on the salivary amylase( s AA) activity and salivary flow rate in rats and give a guideline for saliva collection in response to gustatory stimulus. Methods The unstimulated whole saliva samples in 30 female SD rats were collected. After the tongue was stimulated by citric acid filter paper for 30 s at intervals of 2.5 min,the stimulated saliva were collected at 5,10 and 15 min,respectively. s AA activity was detected by Bernfeld,and the correlation of s AA activity and salivary flow rate was analyzed. Results The s AA activity and salivary flow rate increased significantly,including ratios( P〈0. 05). s AA activity and flow rate among three time points of stimulated samples showed no significant difference( P〈0.05). A significant positive correlation was observed between s AA activity and salivary flow rate in unstimulated situation( P〈0. 05),but a negative correlation was noted under stimulation( P〈0.05). Conclusion Citric acid stimulation may increase the s AA activity and salivary flow rate in rats. This convenient method is suitable for saliva collection.
