摘要
目的研究毛竹笋醇提取物对小鼠酒精性肝损伤的保护作用及初步的机制。方法 60只小鼠随机分成6组,即空白组、模型组、阳性药(联苯双脂)组及毛竹笋醇提取物高、中、低剂量组。除空白组以外,其余组小鼠用75%医用酒精勾兑成的25%、35%、45%三个浓度梯度,以每个浓度梯度灌胃5 d,共15 d复制肝损伤模型;同时,不同给药组灌胃相应的药物,空白组和模型组则灌胃等量双蒸水。观察毛竹笋醇提取物对模型小鼠血清谷草转氨酶(AST)、谷丙转氨酶(ALT)、超氧化物歧化酶(SOD)、丙二醛(MDA)和肝组织细胞形态变化的影响。结果不同浓度的毛竹笋醇提取物均能降低酒精性肝损伤小鼠血清中的ALT,而仅有毛竹笋醇提取物中、低剂量组能降低血清AST。毛竹笋醇提取物高、低剂量组均能升高酒精性肝损伤小鼠血清中SOD且能降低血清MDA。病理组织切片显示,不同浓度的毛竹笋醇提取物均能减轻肝细胞脂肪变性。结论毛竹笋醇提取物对小鼠酒精性肝损伤有保护作用,其作用机制可能与抗氧化损伤相关。
Objective To investigate the effect of ethanol extract of bamboo shoots on acute alcohol - induced hepatic injury in mice and its mechanism. Methods Sixty mice were randomly divided into 6 groups: normal, control, Bifendatatum group, and Bamboo shootg alcohol abstracts with three different concentrations. The medical alcohol(75% ) was blended into 25% ,35%, 45% (v/v) , except sham group, acute hepatic injury was induced by daily administration of three concentration gradient of alco- hol, and each concentration gradient was administrated for 5 days, and all carried out for 15 days, at the same time, normal and control groups were given equal volume of pure water, the other groups administered drugs. The level of serum AST, ALT, SOD, MDA and the histological changes of liver were observed. Results Different concentration Bamboo shoot~ alcohol abstracts could decrease the level of serum ALT in alcohol - induced liver injury model, while medium and lower does of the alcohol abstracts could reduce the level of AST, in addition, higher and lower does of the alcohol abstracts could increase SOD activity and reduce MDA content. The different does of Bamboo shoot's alcohol abstracts reduced the degree of liver steatosis induced by alcohol. Conclusion The ethanol extract of bamboo shoots has significant protection against acute alcohol - induced hepatic injury in mice, and the mechanism may be associated with its antioxidative activities.
