“二神丸”中药物炮制前后对脾肾阳虚泄泻大鼠血清中一氧化氮和胃动素、胃泌素的影响 被引量:7

摘要 目的:比较"二神丸"中两味药物炮制前后对脾肾阳虚泄泻模型大鼠血清中一氧化氮(NO)、胃动素(MTL)、胃泌素(GAS)含量的影响,拟阐明"二神丸"中药物炮制增强疗效作用机理。方法:将50只SD雄性大鼠随机分为正常、模型、阳性药物、生品"二神丸"、炮制品"二神丸"5组,通过复合因素造脾肾阳虚泄泻模型,并给药后取血清,用酶联免疫吸附法(ELISA)测定大鼠血清中一氧化氮、胃动素、胃泌素的水平。结果:模型组血清中一氧化氮、胃动素、胃泌素水平与正常组相比均有显著增高(P〈0.05);三个给药组均能调节三者趋于正常水平,且炮制品"二神丸"组对一氧化氮、胃动素和胃泌素的调节作用比生品"二神丸"组更明显(P〈0.05~0.01)。结论:"二神丸"中的药物炮制后对大鼠血清中一氧化氮、胃动素和胃泌素的调节作用比生品"二神丸"组更为明显,提示复方"二神丸"中药物炮制后增强止泻作用可能是通过纠正异常的一氧化氮水平和消化道激素而达到的。 Objective: To compare the influence of two medicines before and after processing in "Ershen Wan" on the content of NO, motilin(MTL), gastrin(GAS) in serum of Spleen-Kidney Yang deficiency rats, and provide experimental basis for mechanism of enhancing curative effect after processing. Method: 50 SD male rats were randomly divided into 5 groups: normal, model, positive drug, "Ershen Wan", and "Ershen Wan" processed. Spleen-kidney yang deficiency diarrhea rats model were established with composite factors. The plasma from arteria cruralis was collected after treatment, and the contents of NO, MTL, and Gas were detected using ELISA method. Result: NO、MTL、Gas in model group were significantly increased compared with normal group(P〈0.05). The positive, " Ershen Wan" and " Ershen Wan" processed groups can all significantly regulate the contents of NO, MTL, and Gas to normal level. Compared with " Ershen Wan " group, the impact on NO, MTL, and Gas was more significant in " Ershen Wan " processed group(P〈0.05~0.01). Conclusion: The component medicine in "Ershen Wan" after processing can strength antidiarrheal effects which suggest that the mechanism may be related to the mediation by retrieving abnormal levels of NO and gastrointestinal hormone.
