黄药子配伍甘草对大鼠肝脏CYP1A2、CYP2E1蛋白表达影响的实验研究 被引量:6

摘要 目的:研究黄药子和黄药子配伍甘草后对大鼠肝脏组织CYP1A2、CYP2E1蛋白表达的影响。方法:将24只雄性SD大鼠分为3组,分别给药28天,采用HE染色观察肝组织病理损伤,免疫组化法检测大鼠肝脏CYP1A2、CYP2E1蛋白表达的情况。结果:黄药子与甘草配伍能够改善大鼠肝脏组织病变;与对照组比较,黄药子组肝脏CYP1A2、CYP2E1蛋白的阳性表达指数明显升高(P<0.01);与黄药子组比较,配伍组肝脏CYP1A2、CYP2E1蛋白的阳性表达指数降低(P<0.05或P<0.01)。结论:甘草能对抗黄药子导致的肝损伤,其减毒机理可能与抑制肝脏CYP1A2、CYP2E1蛋白表达有关。 Objective: To study the effects of Huangyaozi used alone and combined with Gancao on the protein expression of CYP1A2 and CYP2E1 of the liver in rats. Method: 24 SD rats were randomized into 3 groups. After 28 days of administration of drugs respectively, the pathological alterations were observed after HE staining, and the protein expression of CYP1A2 and CYP2E1 of the liver in rats were detected by immunohistochemistry. Result: The compatibility of Huangyaozi and Gancao can amend the liver tissue of rats with lesions. Compared with the control group, the protein expression of CYP1A2 and CYP2E1 of the liver in the Huangyaozi group were significantly increased(P〈0.01).Compared with the Huangyaozi Group, the protein expression of CYP1A2 and CYP2E1 of the liver in the combination group were decreased(P〈0.05 or P〈0.01). Conclusion: Gancao can lower the hepatic injury induced by Huangyaozi. The mechanism may be related to the inhibition of CYP1A2 and CYP2E1 protein expression.
