老年白内障晶状体上皮细胞中caspase-3、Fas/FasL的表达及意义

Apoptosis and Expression of Caspase-3,Fas,Fas L in Lens Epithelial Cells of the Age-related Cataract

目的:探讨老年白内障晶状体上皮细胞(LECs)中caspase-3、Fas/Fas L的表达及临床意义。方法:选择老年白内障患者25例为研究对象,并收集10例老年人尸体解剖非白内障眼球为对照组。采用免疫组化法检测各组晶状体前囊膜组织中caspase-3、Fas、Fas L的表达。结果:研究组、对照组的LECs中凋亡细胞百分率分别为34.0%、2.0%,研究组凋亡细胞百分率显著高于对照组,差异有统计学意义(P<0.01)。研究组LECs中caspase-3、Fas、Fas L蛋白的阳性表达率分别为84.0%、60.0%、68.0%,而对照组LECs中caspase-3、Fas、Fas L蛋白的阳性表达率为10.0%、10.0%、20.0%,两组caspase-3、Fas、Fas L蛋白阳性表达率比较差异均有统计学意义(P<0.01)。在25例老年性白内障患者中,LECs凋亡与caspase-3和Fas、Fas L表达水平均成正相关的关系(r=0.621、0.583、0.621,P<0.01)。结论:老年性白内障形成过程可能与LECs的凋亡及caspase-3、Fas/Fas L的表达调控有关。

Objective：To investigate the apoptosis and expression of caspase-3,Fas,Fas L in lens epithelial cells of the age-related cataract.Method：25 cases with age-related cataract（study group）and 10 normal controls（control group） were collected.The expressions of caspase-3,Fas,Fas L in LECs were detected by immunohistochemical staining.Result：The apoptosis rates of study group,control group were 34.0%,2.0%.The apoptosis rates of study group were higher than those of control group.The expression of caspase-3,Fas,Fas L in LECs of study group were 84.0%,60.0%,68.0%.The expression of caspase-3,Fas,Fas L in LECs of control group were 10.0%,10.0%,20.0%.There were significant difference in the expression of caspase-3,Fas,Fas L of two groups（P〈0.01）.It showed a positive correlation with the percentage of apoptotic cells（r=0.621,0.583,0.621,P〈0.01）.Conclusion：The apoptosis of LECs and expressions of caspase-3,Fas,Fas L were related to pathogenesis of age-related cataract.
