摘要
目的:建立高效液相色谱-二极管阵列检测法同时测定白花蛇舌草中5个环烯醚萜成分含量的方法,为制定白花蛇舌草质量标准提供参考。方法:采用ZORBAX Extend-C18(250 mm×4.6 mm,5μm)色谱柱;以乙腈(B)-0.05%磷酸溶液(A)梯度洗脱,检测波长238 nm,流速1.0 ml·min^-1,柱温30℃。结果:鸡屎藤次苷甲酯、车叶草酸、车叶草苷、反式6-O-香豆酰鸡屎藤次苷甲酯和反式6-O-阿魏酰鸡屎藤次苷甲酯各自浓度范围内线性关系良好(r〉0.999 6),平均加样回收率为97.20%-100.6%,RSD均小于3%(n=6)。应用所建方法测定了不同批次白花蛇舌草中5个环烯醚萜成分的含量。结论:该方法简单、快速、准确,可为白花蛇舌草药材质量控制提供参考。
OBJECFIVE To develop a method of high performance liquid chromatography coupled with a photodiode array detector for the content determination of scandosidemethyl ester, asperulosidic acid, asperuloside, (E) -6-O-p-coumaroyl scandoside methyl ester and (E) -6-O-feruloyl scandoside methyl ester in Oldenlandia diffusa (Wind.) Roxb. METHODS The analyses were performed on a ZORBAX Extend-C18 column (250 mm × 4. 6 mm, 5 μm) eluted with aeetonitrile (B)-0. 05% phosphoric acid (A) in gradient mode at a flow rate of 1.0 ml·min^-1, a detection wavelength of 238 nm, and a column temperature at 30℃. RESULTS Five iridoids have good linear relationship in their respective concentration range (r〉0. 999 6), the mean recovery rates were 97. 20% - 100. 6%, RSD 〈3 % (n = 6). Five iridoids of different batches of Oldenlandia diffusa (Wind.)Roxb. were determined by using our HPLC-DAD. CONCLUSION The method is simple, rapid and accurate, and can be used as the quality control for Oldenlandia diffusa (Wind.) Roxb..
