电针内关、神门对心肌肥厚模型大鼠AngⅡ和ET影响的比较研究 被引量:7

Comparative study of electro-acupuncture on Neiguan(PC6) and Shenmen(HT6) acupoints influencing AngⅡ and ET in myocardial hypertrophic rats

摘要 目的:观察电针内关、神门穴对异丙肾上腺素(ISO)致心肌肥厚模型大鼠血管紧张素(AngⅡ)和内皮素(ET)的影响,为针灸临床研究方法提供思路。方法:SD雌性大鼠60只,随机分为4组,每组15只:正常组背部皮下注射0.9%氯化钠溶液;模型组每日上午背部皮下注射ISO,3.0mg·kg-1·d-1;电针组分为内关组、神门组,皮下注射ISO 3mg·kg-1·d-1,接HANS-200型电针治疗仪,连续波,频率2Hz,强度1m A,通电20min,1次/d,共14d。所有大鼠于相应处理结束后心电图记录观察并称重隔夜取材:麻醉后开胸摘取心脏并分离左心室称重,放射免疫法检测AngⅡ和ET。结果:1与正常组比较,模型组大鼠ST段显著抬高,心肌肥厚指数升高,AngⅡ和ET含量增高,差异均有统计学意义(P<0.05);2与模型组比较,电针两组大鼠ST段抬高幅度均变小,心肌组织中AngⅡ和ET含量均减少,差异均有统计学意义(P<0.05),两电针组之间比较无统计学差异;内关组、神门组心肌肥厚指数均下降,两组间比较差异无统计学意义。结论:1电针能使心肌肥厚大鼠ST段趋于正常,内关、神门穴作用无统计学差异;2电针能降低心肌肥厚大鼠AngⅡ和ET含量,这可能是电针改善心肌肥厚的神经递质调节作用机制之一。 Objective: To observe the influence of electro-acupuncture on angiotensin Ⅱ (Ang Ⅱ ) and endothelin (ET) of myocardial hypertrophy rat model which was established by injecting isoprenaline (ISO) by acupuncturing on acupoints of Neiguan and Shenmen, to provide thought for clinical research of acupuncture and provide a scientific basis for researches on relations of acupoints and viscera. Methods: 60 female SD rats were randomly divided into 4 groups by random number table method, with 15 rats in each group. Rats in normal control group were treated with injection of physiological saline in the back skin for 14 days; rats in model group were injected with ISO with a dosage of 3mg · kg^-1 · d ^-1 for 14 days; rats in electroacupuncture (EA) groups were received with electro-acupuncture on Neiguan (PC 6) and Shenmen (HT 7) treatment after the ISO injecting, and the stimulus parameter as following: continuous-wave, frequency: 2HZ, current intensity: lmA, and treated for twenty minutes a day, treatment for 14 days. Electrocardiograms (ECG) and weight of all the rats were recorded after the corresponding processing. All the rats were sacrificed on the next day. Rats were anesthetized, then their chest was open to take out hearts and separate the left ventricles. Radioimmunoassay method was used to detect the Ang Ⅱ and ET contents. Then the data were analyzed. Results: (1)Compared with the normal control group, the ECG ST segments, heart rate variability index (HRVI), Ang Ⅱ and ET contents of rats in the model group were significantly higher (P〈0.05). (2)Compared with the model group, the ECG ST segments, Ang Ⅱ and ET contents of rats in EA groups were declined significantly (P〈0.05). There was no significant difference between Neiguan and Shenmen group. Conclusion: (1)EA can make the ST segment of rats with myocardial hypertrophy tend to be normal, and there is no significant difference in effect between Neiguan and Shenmen acupoints. (2)EA can reduce the Ang Ⅱ and ET contents in rats with myocardial hypertrophy, which may be one of the neurotransmitter regulating mechanisms of improving myocardial hypertrophy by EA.
