新加金水六君方对哮喘小鼠气道炎症及气道反应性的影响 被引量:7

Effects of Modified Jinshui Liujun Decoction on the airway inflammation and airway reactivity of the asthmatic mice

摘要 目的:探讨新加金水六君方对哮喘小鼠气道反应性、炎性细胞及肺组织病理的影响。方法:将60只小鼠平均随机分为6组:空白组,模型组,地塞米松组,附子半夏组,金水六君煎组,新加金水六君方组。除空白组外,各组均腹腔注射及雾化吸入卵蛋白(OVA)致敏液,造成慢性哮喘模型。各给药组予灌胃给药共14d。50d测定血液中嗜酸性粒细胞(EOS)计数、炎性细胞百分比以及肺泡灌洗液(BALF)中炎性细胞百分比,观察肺组织病理学改变,并取新加金水六君方组进行气道反应性检测。结果:与模型组比较,新加金水六君方组血液中EOS含量、BALF中EOS百分比均显著降低(P<0.01),病理较其余各组改善最为显著;新加金水六君方高、中、低剂量能够显著降低肺阻力(RL)(P<0.05,P<0.01),显著升高Cdny(P<0.05,P<0.01)。结论:新加金水六君方对哮喘小鼠的气道反应性、炎性细胞和肺组织病理均有明显改善作用,该方中所含十八反附子半夏反药配伍未见影响总体药效水平,由于反药配伍是否产生副作用值得深入探讨。 Objective: To investigate the effects of Modified Jinshui Liujun Decoction on the airway responsiveness, inflammatory cells and pathological changes of lung tissue of the asthmatic mice. Methods: 60 mice were randomly divided into six groups: blank group, model group, dexamethasone group, Fuzi Banxia group, Jinshui Liujun Decoction group, Modified Jinshui Liujun Decocion, with 10 mice in each group. Except the blank group, all the other groups were sensitized by intraperitoneal injection and atomization inhalation with OVA sensitizing solution to set up chronic asthma models. The treatment groups were given intragastric administration. The blood eosinophils, inflammatory cells percentage and inflammatory cells percentage in bronchoalveolar lavage fluid (BALF) were detected on the 50th day, to observe the pathological changes of lung tissue, and select Modified Jinshui Liujun Decoction group to take airway responsiveness test. Results: Compared with model group, the blood eosinophils, the eosinophils percentage of BALF in Modified Jinshui Liujun Decoction group was significantly lower (P〈0.01). Compared with the other groups, the pathological changes of the lung tissue and bronchus were significantly improved in the Modified Jinshui Liujun Decoction group. High, medium and low doses of Modified Jinshui Liujun Decoction groups reduced RL (P〈0.05, P〈0.01), and increased Cdny significantly (P〈0.05, P〈0.01). Conclusion: The Modified Jinshui Liujun Decoction could improve airway responsiveness, inflammatory cells and pathological changes of lung tissue of the asthmatic mice, which was proved to be more effective than Jinshui Liujun Decoction. The incompatible drug pair of radix aconiti lateralis praeparata and rhizoma pineUiae did not influence the overall therapeutic effect, and possible adverse reactions need to be investigated in further study.
