双阴性T细胞与HIV-1疾病进程相关 被引量:1

Double negative T cells correlate with disease progression during HIV-1 infection

摘要 目的探讨双阴性T细胞(double negative T cells,DNT)与人类免疫缺陷病毒(HIV-1)疾病进程相关性。方法 HIV-1感染者(150例)源于北京佑安医院男男性接触者(MSM)高危人群筛查队列。留取患者第一次血清抗体阳转随访点及1、2、4、8、12、24、36、48、60、72、96、108周随访点血液标本。分别用CD3-APC、CD4-FITC和CD8-PE抗体进行细胞表面染色,并通过流式细胞仪检测T细胞及DNT细胞数量。血浆病毒载量通过实时定量PCR方法进行检测。利用SPSS 11.5软件对数据进行分析,P<0.05为差异有统计学意义。结果 DNT细胞的动力学变化与CD4+T细胞的动力学变化一致。DNT细胞数量与CD4+T细胞数量呈正相关,且CD4+T细胞数量较高患者其DNT细胞数量也相应较高。DNT细胞数量与病毒载量呈负相关,且病毒载量较低患者其DNT细胞数量较高。结论DNT细胞的数量与HIV-1疾病进程相关。 Objective To study the dynamics of double negative T cells(DNT)cells and the impact of DNT cells on disease progression of HIV-1infection.Methods HIV-1infected individuals(n=150)were enrolled from MSM high-risk cohort in Beijing You′an Hospital of China.Blood samples were obtained from HIV-1infected individuals at the first antibody seroconversion time point and the 1,2,4,8,12,24,36,48,60,72,96,108 week points thereafter.Whole-blood lyses and T lymphocyte and DNT cell counts were determined by flow cytometry using CD3-APC,CD4-FITC and CD8-PE monoclonal antibodies.Plasma viral load was quantified by real-time PCR.Results The dynamics of DNT cells is similar to that of CD4+T cells.DNT cells positively correlated with CD4+T cell counts and negatively correlated with plasma viral load.DNT cell numbers were higher in the group with high CD4+T cells than that in the group of low CD4+T cells and lower in the group with high viral load than that with low viral load.Conclusions DNT cell counts correlated with HIV-1disease progression.
