大鼠肺鳞癌组织中Pokemon基因与MDM2基因表达的相关性

摘要 目的:探讨原癌基因Pokemon与癌基因MDM2在大鼠肺鳞癌发生发展过程中的表达及意义.方法:采用Wistar大鼠实验组灌注三甲基胆蒽与二乙基亚硝胺诱癌,对照组灌注碘油,采用原位杂交法检测肺鳞癌发生、发展各阶段的基因转录及表达水平.结果:实验组诱癌率为80%,其中支气管黏膜上皮增生28%,不典型增生17.3%,原位癌37.3%,侵袭癌26.7%,转移癌21.3%.Pokemon基因对照组与不典型增生组、鳞癌组比较差异有统计学意义;转移癌组和非转移癌组比较差异有统计学意义.MDM2基因对照组与不典型增生组、鳞癌组比较差异有统计学意义,转移癌与非转移癌比较差异有统计学意义.Pokemon基因与MDM2基因两者表达呈正相关.结论:Pokemon基因与MDM2基因在大鼠肺鳞癌的演进过程中呈现逐渐的高表达,两者可能共同参与了大鼠肺鳞癌的发生、发展过程. Objective:To investigate the expression and significance of Pokemon proto-oncogene and MDM2 oncogene expression in rat lung squamous cell carcinoma.Methods:We established a rat lung squamous cell carcinoma model and used in situ hybridization to semi-quantitatively analyze Pokemon and MDM2 expression in various stages of lung squamous cell carcinoma.Ninety Wistar rats were included,with 15 in the control group and 75 in the experimental group.Results:The rate of cancer induction was 80% (60/75),that of bronchial epithelial hyperplasia was 28%,that of atypical hyperplasia was 17.3 %,that of carcinoma in situ was 37.3 %,that of invasive carcinoma was 26.7 %,that of metastatic carcinoma was 21.3 %.We found a significant difference in Pokemon expression in the control group compared with the atypical hyperplasia and squamous cell carcinoma groups.There was a significant difference in MDM2 expression in the control group compared with the atypical hyperplasia,squamous cell carcinoma groups.The differences in expression of both genes in the metastasis and non-metastasis group were statistically significant.Pokemon gene expression was positively correlated with MDM2 gene expression.Conclusion:The genes of Pokemon and MDM2 show gradual rising expressions during the evolution of rat lung squamous cell carcinoma.And the two genes might be involved in the carcinogenesis of rat lung squamous cell carcinoma.
