摘要
为克服传统繁殖方法繁殖系数低和周期长等缺点,实现龙芽草的快速繁殖,以龙芽草(Agrimonia pilosa)叶片为外植体和MS为基本培养基,利用不同生长调节物质及其组合,研究建立了龙芽草组织培养的快速繁殖体系。结果表明:细胞分裂素可直接诱导叶片产生不定芽,MS+TDZ 0.3 mg/L+6-BA2.0mg/L为适宜培养基;增殖生长需添加生长素,适宜的培养基为MS+TDZ 0.3 mg/L+6-BA2.0mg/L+IAA 1.0mg/L;肌醇不影响生根,适宜生根的培养基为1/4 MS+NAA 0.1mg/L。
To overcome the shortcomings of low propagation coefficients and long cyclicality,and realize the rapid propagation of A.pilosa,leaf and MS were use as explants and basic medium separately to construct rapid propagation system by using different growth regulatory substances and their combinations.The results showed cytokinin could induce adventitious buds.The optimal medium for adventitious buds was MS+TDZ 0.3mg/L+6-BA 2.0mg/L;Auxin was needed for proliferation culture,the optimal medium for proliferation was MS+TDZ 0.3 mg/L+6-BA 2.0 mg/L+IAA 1.0 mg/L;Rooting culture test showed that inositol had no effect on rooting.So the optimal medium for rooting culture was 1/4MS+NAA 0.1mg/L.
