摘要
东方田鼠具有天然抗日本血吸虫活性,为了筛选抗日本血吸虫疫苗分子,通过日本血吸虫尾蚴感染东方田鼠和昆明小鼠,感染11天后收集东方田鼠和昆明小鼠肝童虫.肝童虫经组织匀浆收集蛋白质并进行蛋白质定量后SDS-PAGE分离,切取差异蛋白条带进行蛋白质鉴定.发现差异蛋白主要为日本血吸虫结构蛋白,能量代谢相关蛋白以及热休克蛋白等.为进一步研究抗日本血吸虫疫苗奠定了基础.
Microtus fortis is naturally resistant to Schistosoma japonicum. In order to find against Schistosoma japonicum antigen, the Microtus fortis and Kunming mouse are used in this study. They are challenged with cercariae of Schistosoma japonicum. The livers of the microtus fortis and mouse are collected at 11 days after infection. The proteins from the hepatic Schistosomula separated with SDS-PAGE. The differential proteins are analyzed by in-gel digestion and MS/MS identification. The result shows that the structure protein and energy metabolism protein are significantly down-regulated in the liver of Microtus fortis at 11 days after infection. The heat shock proteins are significantly up-regulated in the liver of Mierotus fortis at 11 days after infection.
