摘要
为确立一种耗时短、结果清晰的筛选产纤维素酶丝状真菌的平板法,比较了不同染色方式、不同培养基碳源的三种刚果红法的效果。结果表明,以纤维素粉和葡萄糖为复合碳源的培养基培养至菌落形成后,再以刚果红染液染色的方法最好,菌丝生长快、形成的透明圈边缘清晰,直观性强。
In order to get an optimum congo red-staining method for the detection of cellulose-producing filamentous fungi by way of plate assay, three methods of different staining way and medium with different carbon source were comparcd. The results showed that the third one was best. In this method, mycelial mass of microorganisms were inoculated on the medium which contains microcrystalline cellulose and glucose as the carbon sources. After a cultivation period, the plates were flooded with Congo red. The method is rapid and efficient. Filamentous fungi grow rapidly and transparent zone formed around the cellulase-producing microbial colonies was clear.
