促红细胞生成素对大鼠脑缺血再灌注后血脑屏障通透性和ZO-1表达的影响 被引量:3

摘要 目的研究促红细胞生成素(erythropoietin,EPO)预处理对大鼠脑缺血再灌注后血脑屏障通透性和紧密连接蛋白ZO-1的影响。方法采用大鼠大脑中动脉栓塞(MCAO)模型,伊文思蓝渗透性实验检测血脑屏障通透性,免疫组化法和Western blot法测定ZO-1蛋白表达,RT-PCR法测定ZO-1 mRNA表达。结果 EPO预处理能够明显减轻脑缺血再灌注后血脑屏障的通透性(P<0.01),并显著上调缺血脑组织中ZO-1蛋白及mRNA的表达水平(P<0.01)。结论 EPO通过上调脑缺血再灌注后脑组织中ZO-1的表达,降低BBB通透性,这可能是EPO保护脑缺血再灌注后神经组织的机制之一。 Objective To study the effects of erythropoietin (EPO) pretreatment on blood brain barrier (BBB) and ZO-1 expression after cerebral ischemia-reperfusion in rats. Methods Using middle cerebral artery occlusion (MCAO) model in rats, the BBB permeability was assessed by Evans blue extravasation. The changes of ZO-1 protein and mRNA were determined by immunohistochemistry assays, Western blot, and reverse transcriptase-polymerase chain reaction, respectively. Results EPO significantly reduced BBB permeability(P 〈 0.01 ) and markedly increased the expressions of ZO-1 protein and mRNA in ischemia tissue after cerebral ischemia-reperfusion ( P 〈 0.01 ). Conclusion EPO could decrease the BBB permeability by increasing the expression of ZO-1 ,and which may be one of the protection mechanism of EPO on brain tissue after cerebral ischemia-reperfusion.
