摘要 以杉木( Cunninghina lanceolata)幼苗茎段为外植体,研究升汞( HgCl2)和次氯酸钠( NaClO)灭菌在杉木组织培养过程中对外植体污染以及不定芽萌发的影响。结果表明:升汞对杉木茎段的灭菌效果优于次氯酸钠,0.1%升汞灭菌11-15 min无菌活体的获得率可达66.7%以上;用5%次氯酸钠灭菌后的无菌活体的获得率最高仅为26.7%,且延长的灭菌时间,对外植体污染率及灭菌成功率影响不大。但与次氯酸钠相比,升汞灭菌对不定芽萌发的负面影响较大,其不定芽萌发时间晚,出芽率及出芽指数相对较小。 The tender stems from Cunninghina lanceolata were used as explants to study the effects of sterilization of mercuric chloride ( HgCl2 ) and chloros ( NaClO) on pollution of explants and germination of adventitious bud in Cunninghina lanceolata tissue culture. The results showed that the sterilizing effect of HgCl2 on the stems of Cunninghina lanceolata was better than that of NaClO. The pick-up rate of living aseptic reached above 66. 7% with 0. 1% HgCl2 treatment from 11 minutes to 15 minutes. The highest pick-up rate of living aseptic was 26. 7% with 5% NaClO treatment, the effect of which on the pollution rate and the success rate of sterilization was not obvious if extending the sterilization time. Compared with HgCl2 , the negative effect of NaClO treatment on adventitious bud was stronger. Germination time of ad-ventitious bud was late, and the budding rate and budding index was relatively small.
